Cloning and Expression Analysis of GhPR -10 Gene Related to Drought Stress in Gossypium hirsutum

Abstract:The generation and accumu lation of pathogenesis-related proteins (PR) is one of the prominent features of plant responding to abiotic or non-abiotic stress, and pathogenesis-related protein 10 is an important memberof this kind of protein family. Based on the preliminary proteomic analysis, the study designed a pair of primers by combining homology cloning approaches with RACE (rapid-amplification of cDNA ends) techniques and cloned a differentially expressed gene in Gossypium hirsutum "KK1543"under drought stress and it was named GhPR -10. The gene had an open reading frame of 432 bp and encoded 144 amino acids. Predicted molecular weight was about 15.6 kD and isoelectric point was 4.90. Amino acid sequence alignment and homology analysis revealed GhPR-10 protein and PR-10 protein in other plants had higher consistency, which indicated that the gene in the process of evolution was quite conservative. Phylogenetic tree analysis showed that GhPR-10 protein in Gossypium hirsutum and the proteins in Gossypium barbadense, Gossypium raimondii had the closest relationship. The fluorescence quantitative PCR technology was applied to investigate the expression of this gene in different tissues and organs under different stresses. Analysis results showed that under 15% PEG stress at the same time, the expression of GhPR-10 gene in the roots was higher than that in the stems and leaves. The variation trend of GhPR-10 expression was similar under three stress and all showed an up-regulated expression. The expression level of the gene reached the highest at 24 h under drought and salt stress and it reached the highest at 12 h under 4℃low temperature stress. In conclusion, GhPR-10 genes might be involved in the the mechanism of cotton resistance to stress, and laid a foundation for further research on the function of GhPR-10 gene.
