Abstract:Xanthomonas campestris pv.campestris (Xcc),a yellow pigmented γ-proteobacterium,is the causal agent of black rot disease,one of the major cruciferous plant diseases worldwide.The bacterium harbors a 41.7 kb Hrp pathogenicity island (Hrp PAI) consisting of 41 open reading frames (ORFs),bordered by the IS1479 at 5′ end and IS1595 at 3′ end. The G+C content of the Hrp PAI is 62.3%,lower than that of the whole gnome,64.9%. Hrp PAI region has a tripartite mosaic structure composed of a hrp cluster flanked by a hypothetical gene region (HGR) on the left and an catabolic enzyme gene region (EGR) on the right.The hrp cluster is conserved in Xanthomonas genus,whereas the HGR and the EGR are relatively changeable.The HGR is about 7.4 kb in length and consists of 6 ORFs,all predicted as hypothetical genes,and the EGR is about 9.3 kb and consists of 8 ORFs,of which 6 ORFs are predicted as catabolic enzyme genes. For function characterization,the deletion mutant of EGR of Xcc 8004 was constructed by using marker exchange method. The mutant 8004ΔR was attenuated in virulence on host plant Chinese radish but retained its ability to cause the hypersensitive response in non host plant pepper ECW-10R.No significant differences were found in the production of exopolysaccharide or the activity of extracellular enzymes between the mutant andthe wild type strain.The results showed that the deletion mutation had no effects on the function oftype Ⅲ secretion system (TTSS) and the bacterial growth in minimum medium,but resulted in a significant reduction in the bacterial virulence,suggesting that the right flanking region of hrp cluster of Xcc 8004 might harbor some pathogenicity-related genes.
