Inducement of Philippine Ground Orchid (spathoglottis Plicata) Protocorm-Like Bodies and Plant Regeneration

Abstract:In order to establish a regeneration system in Spcthoglottis plicata,its axillary bud was used as explant to study the effect of different hormone concentration and additives on PLBs induction,differentiation and rooting.The results showed that sterile plantlets were obtained under the condition of 1/2 MS+2.0 mg/L 6-BA+0.2 mg/L NAA+0.2 g/L AC (activated carbon)+10％,CW (coconut juice) after 35 days and its survival rate was higher that 80％.The PLBs induction of apical meristem reached its top under the condition of 10 mg/L Picloram+l/2 MS,and the highest induction rate of PLBs clusters was 75％.Plantlets were differentiated from the PLBs under the condition of 1/2 MS+2.0 mg/L 6-BA+0.2 mg/L IBA after 45 d and the differentiation rate was 54％.The regeneration rate could reach up to 85％ under the condition of 1.0 mg/L IBA+0.5 mg/L 6-BA+0.2 g/L AC after 35 days.The survival rate of transplanted seedlings reached 95％ with excellent growth.
