Secretory expression of mSTa, LTB and STb fusion protein of ETEC in Lactococcus lactis and the immunogenicity analysis

Abstract:To evaluate the immune responses of recombinant L.lactis NZ9000 expressing ETEC STa,LTb and STb fusion protein as oral vaccine,the fusion gene of the promoter p23,signal peptide USP45,and ETEC mSTa-LTb-STb was cloned into the expression vector pTX8048 in turn,and the expression vector pTX-sls was constnuted.Then the pTX-sls was transformed into L.lactis NZ9000 by electroporation,resulting in recombinant strain pTX-sls/L.lactis NZ9000.The interest proteins expressed by the recombinant strains were detected by western blot and indirect ELISA.BALB/c mice were used as animal model immunized with recombinant strains by intragastric administration,and the immune efficacy was analyzed.The results showed the expressed protein possessed the antigenic specificity,and secreted into the culture supematant.The results of ELISA showed that the mice immunized with recombinant strains could produce remarkable specific sIgA level in the feces,and high level of anti-LTB protein IgG in the serum (p<0.01).Statistical significant difference was observed among the spleen lymphocyte proliferation index (LPI) among the immunization groups of mice and control groups.The induced antibodies in serum demonstrated neutralizing effect on STb infection.In conclusion,the oral immunizations with recombinant L.lactis NZ9000 could induce significantly specific mucosal IgA response as well as serum IgG responses,and could evoke both mucosal immune and system immune responses.Our data indicated that intragastric administration of the recombinant L.lactis NZ9000 could induce specific immune response against ETEC,and established a good foundation for further study on the new and effective ETEC oral vaccines.
