Alternative Splicing, Multiple Transcription Initiation Sites of Nicotinic Acetylcholine Receptor Subunits from the Cotton Aphid Aphis Gossypii

Abstract:Nicotinic acetylcholine receptor (nAChR) plays important roles in neural transmission and is one of only a few validated target sites for pesticides. Three nAChR subunits (Agα1, Agα2 and Agβ1) were cloned from Aphis gossypii with RT-PCR and RACE strategy. Both 5' and 3' cDNA ends of 3 subunits were successfully determined with 5' and 3'-RACE techniques. The 5'-RACE experiments identified three isoforms of Agβ1 subunits, which differ in their 5'-untranlasted regions, suggesting three alternative transcription initiation sites. In addition, the shortest isoform, Agβ1C, undergoes alternative splicing in protein coding regions. A fragment of 186 bp located in loop D region is skipped, which involves in forming the acetylcholine-binding site in heterodimeric receptors. The 3'-RACE experiments were performed to amplified the 3'-end of three subunits' mRNA. In Agα1 subunit, poly (A) was sequenced at the 3'-end, indicating the full length of mRNA. However, it lacks transmembrane domain 4, a conserved and essential domain for the function of the α subunit. Here, we report the complexity of insect nAChR subunit from the cotton aphid, attributing to multiple transcription initiation sites and alternative splicing. In addition, phylogenetic analysis indicated that insect nAChR subunits should be classified into three subgroups, αⅠ, αⅡand β, rather than two as in previous reports. These results indicated that insect nAChR subunits might be more complex than expected, though they are constrained to the neural system
