Effect of Cell Geometry on YAP Localization of Mesenchymal Stem Cells on Micropatterned Surfaces

Abstract:Mechanical forces such as substrate rigidity or geometrical constraints have been proved to direct gene expression and cell differentiation. There is growing evidence that YAP (yes-associated protein) plays an important role in mechanosensing via transmitting through the cytoskeleton and to the nucleus. In this study, we use micropatterned surfaces to manipulate cellular geometries of triangle, square, pentagon, hexagon, circle, diamond and trapezoid with the same small area. Mesenchymal stem cells (MSCs) were cultured on the micropatterned surface and the role of single-cell morphology in the regulation of YAP location was investigated. The results show that geometric constraints decreased YAP nuclear accumulation. In addition, we found that YAP was mostly localized in nuclei at cells with strong actomyosin contractility and in the cytoplasm at cells with thin and less evident stress fibers. Taken together, our work further demonstrates the regulation of YAP cytoplasm/nuclear translocation based on the actomyosin contractility state and the patency of the actin cytoskeleton by single-cell constraints on seven different geometric micropatterns with the same surface area. Actin cytoskeleton may act as a molecular switch in regulation of YAP activity.
