Co-delivery of Paclitaxel and Gene via Chitosan-Derivative-Based Nanoparticles for Enhanced Antitumor Efficacy

Abstract:Nanoparticle-based co-delivery systems have been developed as a promising strategy to deliver chemotherapeutics and genetic drugs.In this study,amphiphilic oleic acid and trimethyl double grafted chitosan nanoparticles(OTC NPs)and folate(FA)modified OTC NPs(FOTC NPs)were prepared for simultaneous delivery of paclitaxel(PTX)and Survivin shRNA-expressing plasmid (iSur-pDNA).Compared to OTC NPs, FOTC NPs possessed slower PTX release,faster pDNA release and higher cellular uptake levels,strongly suppressed the expression of Survivn,greatly facilitated the growth inhibitory effect of PTX,significantly enhanced the in vivo antitumor efficacy.These results indicated that amphiphilic FOTC NPs could serve as an efficient approach for the combinatorial delivery of chemotherapeutic drugs and genes.
