MiR-106 b Regulates Proliferation and Apoptosis of Non-small Cell Lung Cancer by Targeting BTG3

Abstract:Objective To investigate the effect of MiR-106b on the proliferation and apoptosis of non-small cell lung cancer (NSCLC) by targeting BTG3 and its mechanism. Methods qPCR was used to detect the expression of miR-106b in non-small cell lung cancer and adjacent normal tissues. Dual luciferase reporter assay detects the interaction of miR-106b and BTG3;The effect of miR-106b on the proliferation of non-small cell lung cancer cells was examined by MTT cell proliferation assay. The effect of miR-106b on the apoptosis of non-small cell lung cancer cells was detected by flow cytometry. The invasion ability of non-small cell lung cancer cells was detected by Transwell invasion assay, and the expression of BTG3 in tumors derived from nude mice was detected by immunohistochemistry. Results The expression of miR-106b in non-small cell lung cancer tissues was significantly higher than that in adjacent non-small cell lung cancer tissues and the highest expression level of miR-106b in non-small cell lung cancer A549 cells. miR-106b specifically binds to 3'UTR of BTG3 and regulates the expression of BTG3, and inhibits the proliferation and invasion of non-small cell lung cancer cells after miR-106b inhibition. After inhibiting the expres-sion of miR-106b, the tumor volume and weight of the tumor-bearing mice were significantly reduced, and the expression of BTG3 protein was correspondingly reduced. Conclusion miR-106b plays an important role in carcinogenesis and progression of non-small cell lung cancer and may affect the proliferation and apoptosis of non-small cell lung cancer cells by targeting the ex-pression of BTG3.
