Effects of sub-chronic exposure to BPA on ovarian reserve function in female mice

Abstract:Objective To investigate whether sub-chronic oral exposure to low-dose BPA can affect ovarian reserve in female mice.Methods Sixty-four 5-week-old SPF C57BL/6 mice were randomly divided into 4 groups,exposed to 5,50,and 500ug/ml of BPA solution,respectively,and the control group was fed with corn oil alone.After 28 days,blood samples were drawn from the orbital vein of each mice,and the AMH and E2 hormone concentration in serum was detected by ELISA kit.Mice were sacrificed by cervical dislocation,and ovaries were removed immediately.The expression of AMH mRNA was detected by real-time quantitative PCR.The expression of AMH protein was detected by immunohistochemistry.Results Compared with the control group,the level AMH and E2,the expression of AMH gene and protein in the BPA treatment group showed a downward trend.The low dose group and high dose group decreased significantly (P < 0.05).Conclusion There was no single dose effect of BPA on mouse ovary,and sub-chronic exposure to low dose of BPA could induce the decrease of ovarian reserve function in female mice.
