Detection of SLC25A13 gene mutation in a family with primary carnitine deficiency and prenatal diagnosis

Abstract:Objective:To analyze the SLC25A13 gene mutation of Primary carnitine deficiency in a family and provide prenatal genetic diagnosis for this family.Methods:Sanger sequencing was used to screen potential mutations of SLC25A13 in genomic DNA extracted from blood sample of proband and the parents.The amniotic fluid was extracted from the proband's mother for prenatal diagnosis.STR was used to identify the pollution from proband's mother blood to avoid the misdiagnosis caused by pollution of proband's mother blood.Results:Sanger sequencing indicated that the proband has carried two heterozygous mutations of SLC25A13 gene:IVS6+5G>A、c.851de14,which came from his father and mother respectively.The mutation was not detected in healthy members.In amniotic fluid,there were no mutation of these two sites.So the fetus may have different clinical phenotype with proband.Conclusion:The two of SLC25A13 IVS6+5G>A、c.85 ldel4 mutations are hot-spot mutations resultting in NICCD.The Sanger sequencing technology is able to discover the gene mutation in NICCD family,which contributes to genetic counseling and prenatal diagnosis for the NICCD family.
