Protective Effect of Lycium ruthenicum Murr. on Acute Alcoholic Liver Injury Mice and Its Antioxidant Activity

Abstract:Objective To investigate the protective effect of Lycium ruthenicum Murr.(LRM)on acute alcoholic liver injury mice and to observe its antioxidant activity. Methods Fifty male Kunming mice were randomly divided into 5 groups, namely normal control group, model group, and high- dose, middle-dose, low-dose LRM groups(gastric gavage of LRM 2.0, 1.0, 0.5 g·kg-1 for 4 continuous weeks). At the end of 2nd week, the mice were given 56 %(V/V)0.01 L·kg-1 wine,and at the end of 4 th week,the mice were given 0.16 L·kg-1 wine. The serum aspartate aminotransferase(AST)and alanine aminotransferase(ALT)contents, hepatic malondialdehyde(MDA)content,and hepatic reduced glutathione(GSH), superoxide dismutase (SOD), and glutathione peroxidase(GSH-PX) activities were determined. Liver histopathological changes were revealed by HE staining. Results Compared with the model group, LRM could significantly reduce the serum levels of ALT and AST, decrease hepatic MDA content, increase hepatic SOD,GSH and GSH-PX activities(P< 0.05 or P<0.01)in a dose-dependent manner. And LRM could also alleviate the histopathological changes induced by alcohol. Conclusion LRM has protective effects on acute alcoholic liver injury in mice ,and the mechanism is probably related with the improvement of endogenous antioxidant activity of the mice.
