Expression of chemokine receptor CXCR4 in renal cancer patientsand its regulatory mechanisms

Abstract:Objective To study the expression of chemokine receptor CXCR4 in the tumor lesions of renal cancer patients and its possible regulatory mechanisms. Methods The tumor and adjacent tissues were collected and RNA was then extracted. RT-PCR was conducted to detect the expression of CXCR4 in tumor and adjacent tissues. The mRNA expression of transforming growth factor β (TGF-β) in human renal proximal tubular epithelial cell line HKC, and renal cancer cell lines, 769P and 786O were examined using real-time RT-PCR. Replication deficiency adenovirus Ad(E1-).sT and its control Ad(E1-).Null were transfected into 786O cells. TGF-β were added 24 h later for stimulation, and the expression of CXCR4 were detected using real-time RT-PCR. In addition, 24 h after adenoviruses infection, the ability of cell migration were observed by wound healing examination. Results The expression of CXCR4 is much higher in the tumor lesions than that in the para-cancerous tissues. Among total 12 selected patients, CXCR4 was up-regulated in the tumor lesions of 8 patients. Moreover, compared with human renal proximal tubular epithelial cells, the expression of TGF-β, upstream regulator of CXCR4, was significantly increased. In 786O cells, stimulation with TGF-β could up-regulate the expression of CXCR4. Furthermore, Ad(E1-).sT not only inhibit TGF-β-induced CXCR4 expression, but also decreased cellular migration ability. Conclusion CXCR4 expression is increased in tumor lesions of most renal cancer patients, and is regulated by TGF-β. Our results suggest that CXCR4 is a promising target for the therapy of renal cancer.
