Post-traumatic Stress Disorder on the Microcirculation of Mesentery and Cerebral Pia Mater in Rats

Abstract:Objective:To investigate the effect of post-traumatic stress disorder (PTSD)on microcirculation of mesentery and cerebral pia mater in rats and the underlying mechanisms.Method:Sprague-Dawley (SD)rats were randomly divided into two groups:control group (n=6)and PTSD group (n=12).The PTSD model was rep-licated by using the single prolonged stress method.On the 14th day post-stress,the changes of animals'behavior was evaluated via open-field;the microcirculation of mesentery and cerebral pia mater of rats was observed using the microscope;the plasma noradrenaline (NA),endothelial nitric oxide synthase (eNOS)and endothelin-1 (ET-1 ) were measured by using enzyme linked immunosorbent assay (ELISA).Results:Compared with the control rats,the numbers of crossing and rearing of the PTSD rats were decreased significantly in the open-field (P <0.01);the di-ameters of arteriole and veinule in the mesentery and cerebral pia mater from the PTSD rats were significantly thin-ner (P <0.05),the blood flow velocity showed the trend of slowed down;the plasma levels of NA and ET-1 were elevated significantly (P <0.01);there was no significant difference in the plasma level of eNOS (P >0.05).Con-clusion:PTSD induces microcirculation dysfunction in mesentery and cerebral pia mater in rats.
