Relationship between expression of Tiam 1 and invasion and metastasis of gastric cancer

Abstract:Purpose To investigate the expression of T lymphoma invasion and metastasis inducing factor 1 (Tiam1) in tissues of normal gastric mucosa and gastric cancer, also its expression in gastric cancer cell lines, and to analyze the relationship between its expression and invasion and metastasis of gastric cancer. Methods The expression of Tiam1 protein in 60 cases of gastric cancer and their corresponding normal gastric mucosa was detected using strept avidin-biotin complex (SABC) immunohistochemical method. Simultaneously its relationship with the clinicopathological parameters of gastric cancer was also analyzed. RT-PCR and SABC immunohistochemical technology were also applied to detect the expression of Tiam1 mRNA and protein in gastric cancer cell lines, MKN-45 (M_O), its sublines with higher invasion and metastasis potentials (M_H) or with lower invasion and metastasis potentials (M_L). In addition, its relationship with the invasion and metastasis potentials of gastric cancer cells was analyzed. Results There was negative staining of Tiam1 protein in normal gastric mucosa, while the positive rate of Tiam1 protein in gastric cancer was 78.3% (47/60) (P 0.01). With differentiated degree of histologic type descending, depth of invasion increasing, stage of TNM upgrading and lymph node metastasis happening, the positive staining rate of Tiam1 protein in gastric cancer significantly ascended (P0.05), but there was no obvious correlation between its expression and the sex, age, location or size of gastric cancer (P0.05). The expression of Tiam1 mRNA and protein in M_H was much higher than that in M_O and M_L (P0.05), but there was no obvious difference between M_O and M_L (P0.05). Conclusions Positive correlation exists between the expression of Tiam1 and invasion and metastasis of gastric cancer, which suggests Tiam1 may be acted as a new marker indicating its malignant biological behavior.
