Catenin Expression in Human and Rat Liver Tissues and Its Correlation

Abstract:Purpose To investigate catenin expression in human and rat liver tissues and its correlation,providing new evidence for the clinical study of hepatocellular carcinoma. Methods A hepatocellular carcinoma animal model was established by using Walker 256 human liver cancer cell line and Wistar rat. The expression of E-cadherin,α-,β-,γ-and p120 catenin in both human and rat tissues had been examined using immunohistochemical technique. Serum soluble E-cadherin concentration in patient and rat was detected using ELISA method. Results In normal human and rat liver tissues,E-cadherin,α-,β-,γ- and p120 catenin expressed at cell-cell contact region and submembrane,while there were noticeable changes of these protein expression in carcinoma tissues: the membranous expression reduced or lost,and their cytosolic expression increased,β-and p120 catenin had translocated to nucleus (nucleus staining). Serum E-cadherin concentration increased in hepatocellular carcinoma patient and rat as compared with the controls (P0.05). The soluble E-cadherin in BEL-7404 human liver cancer cell line culture medium was higher than that of LO2 normal liver cell,but no significant difference. Conclusions There is a similar expression pattern of catenin family in rat and human hepatocellular carcinoma tissues. Rat liver cancer model system is suitable for the study of the relationship between catenin and hepatocellular carcinoma. The increased E-cadherin in serum and liver cancer cell culture medium is important evidence in liver cancer and is a useful indicator in clinical diagnosis,while the mechanism of the phenomenon needs to be further studied.
