Effects of Hesperetin on Proliferation and Apoptosis of Pancreatic Cancer Panc-1 Cells and Its Mechanism

Abstract:Objective Hesperetin is a natural plant monomer with multiple biological activity, which exerts anti-tumor ability by regulating cell proliferation, apoptosis, metastasis and drug-re-sistance. In this study, we aimed to investigate whether hesperetin could inhibit proliferation and in-duce apoptosis in pancreatic cancer cells and explore the potential mechanism. Methods Pancreat-ic cancer Panc-1 cell line was treated with different concentrations of hesperetin. CCK-8 assay was used to detect cell viability. Flow cytometry was used to detect cell cycle and cell apoptosis. Apop-totic cells, reactive oxygen species ( ROS) and mitochondrial membrane potential were detected by Hoechst 33258, DCFH-DA, and JC-1, respectively. The protein expression levels of AIF, Cyt C, Apaf-1 , Cleaved caspase-9 , BAX and others were detected by Western blotting. Results CCK-8 assay showed hesperetin inhibited proliferation of Panc-1 cells in a time-dependent and dose-depend-ent manner. There were more apoptotic cells in hesperetin-treated group than in the control group ( P<0. 05 ) , determined by Hoechst 3328 staining and FITC-Annexin V/ PI staining. Flow cytom-etry showed that hesperetin induced cell cycle arrest at G0/G1 phase. There was a significant in-crease of ROS and a tremendous loss of mitochondrial membrane potential in hesperetin-treated cells. Western blotting showed that the expression levels of AIF, Cyt C, Apaf-1, Cleaved caspase-9 and BAX increased, while Cyclin D1 decreased. Conclusion Our study demonstrates that hesp-eretin could inhibit cell proliferation and induce apoptosis in pancreatic cancer cells via mitochondri-al-mediated intrinsic pathway by accumulating ROS.
