Abstract:Aim: To observe the influence of Yin-nourishing, Qi-supplementing and Blood circulation-activating recipe on hemorheology and platelel aggregation in rats. Methods: The experiment was conducted at the Anima l Laboratory, Zhejiang College of Traditional Chinese Medicine from February 2003 to August 2004. Forty white rats were selected, and assigned randomly into saline group, aspirin group and 0.5 g/kg Naoshuantong and 2 g/kg Naoshuantong group with 10 in each group. The rats in the aspirin group were treated with 30 mg/kg aspirin with gastric perfusion. Those in the 0.5 g/kg Naoshuantong and 2 g/kg Naoshuantong group were treated with 0.5 g/kg and 2 g/kg Naoshuantong preparation (mainly composed of rehmannia dride rhizome, szechwan lovge rhizome and membranous milkvetch root, etc.) with gastric perfusion, respectively. Those in the control group were treated with saline at the same volume. After continuously giving drugs for 3 days, the blood was gained from carotid artery. Hemorheology index (blood specific viscosity and HCT) and rate of platelet aggregation (1 minute maximal clustering speed, 5 minutes maximal clustering speed) were detected. Results: Among the forty inclu ded white rats, 32 were involved in the result analysis. Other 8 rats were used as compensation of operation. 1 Effects on hemorheology index: The blood specific viscosity in the 2 g/ kg Naoshuantong group at the high cutting speed (230/s) was lower than that in the saline group [5.0±0.4], (5.8±0.5)mPa·s, P < 0.05]. The blood speck viscosity in the aspirin group and 0.5 g /kg and 2 g/kg Naoshuantong group at the low cutting speed (5.75/s) was lower than that in the saline group [(27.6±3.2), (24.0±4.3), (15.6±1.4), (32.5±4.1)mPa·s, P < 0.05;P < 0.01]. 2 Effects on platelet aggregation: The 1-minute maximal clustering speed and 5-minute maximal clustering speed were both lower in the aspirin group and 0.5 g/kg and 2 g/kg Naoshuantong group than those in the saline group [(10.65±6.30),(11.40±2.96),(8.96 ±2.78),(23.01±2.45)%,P < 0.01;(11.78±4.32),(24.13±8.40),(14.06±5.38), (35.42±6.50)%,P < 0.01;P < 0.05]. Conclusion: Yin-nourishing, Qi-supplementing and Blood Circulation-activating recipe has significant effects on reducing blood specific viscosity and HCT, and obviously inhibits the platelet aggregation in vivo in rats induced by adenosine diphosphate. There is markedly dosage-effect positive correlation in Naoshuantong group; especially there are significantly ameliorative effects on every index of hemorheology and platelet aggregation in the big dosage group.
