Transplantation of Endothelial Progenitor Cells from Peripheral Blood Enhancing Neovascularization in Ischemic Tissue of Nude Mice

Abstract:Objective To investigate the impact of endothelial progenitor cells from human peripheral blood on neovascularization in ischemic hindlimb of nude mice.Methods Peripheral blood mononuclear cells from human volunteers were cultured in special cocktail that facilitates growth of EPCs-plating on fibronectin.After7days in culture,1×10 6 EPCs were injected into the tail vein of hindlimb of athymic nude mice model.The animals were randomly separated into EPC group and control group.Histological and physiological evaluation of the transplanted animals was performed after28days.Results Necropsy examination disclosed that capillary density was signifiˉcantly greater in the EPCs group(136.4±40.5/field,P0.01)than that in the control group(53.65±14.02/field)And the rate of limb necrosis and auto_amputation was significantly reduced in the EPCs group.Conclusions Transplantation of Ex vivo expanded EPCs can promote neovescularization in ischemic tissue,incorporate into sites of neovascularization and reduce the rate of limb loss.
