Abstract:OBJECTIVE: With the study progress of proteomics in liver, we review the whole level of hepatic cells, analysis on the proteomics in the level of cell organ, expression difference of liver proteome as well as the problems existing and prospects. DATA SOURCES: We searched PUBMED for the papers published from January 1991 to January 2006, with the of "liver, proteomics, tissue engineering", and the language was limited to English. STUDY SELECTION: The materials were selected primarily and references of each article were looker for. Inclusion criteria: related to hepatic cells and proteomics. Exclusion criteria: repetitive study or Meta analysis. DATA EXTRACTION: Totally 222 related articles were collected, 36 of them met the inclusion criteria, and 186 were excluded due to out-of-date or repetitive content. Among the 18 articles, which met the inclusion criteria, 18 were related to the whole level of hepatic cells, 6 to proteomics analysis on the cell organ level and 12 to expression difference of proteomics. DATA SYNTHESIS: Proteomics is the whole protein expression in some cells, tissue or organs under certain time and condition. Study progress in liver with the points of proteomics is fast. We reviewed the latest progress in the proteomics in protein level of liver. With the two-dimensional gel electrophoresis and mass spectrum technique combined with the cell culture and cell organ separation and other latest experimental techniques, we respectively analysis and confirm the related protein from the level of cells and cell organ, trace some proteins, identify the sorts and physiology of proteins; At the same time, we also studyed the different protein expressions under different pathophysiological status of liver and before and after application of multiple drugs , looked for and determined difference expression protein, which provided help for etiopathogenisis, progression of diseases and drug screening . Progress in hepatic tissue engineering study starting at the 1990s is also very fast, but some problems need to be solved due to the complex hepatic metabolic function, at present such as hepatic seed cell, scaffold, cell culture, vascular ingrowth after implanting, the excretion of hepatic cell metabolism, formation of biliary tract and so on. CONCLUSION: Disease-related protein of difference expression can be used as the target of new drug, also as the marker of disease appearing. Liver is the main part of toxic substance metabolism. Application of high-resolution two -dimensional electrophoresis and mass spectrum is an effective means in observing the expression of different proteins after drug application. The development of tissue engineering depends on the latest achievement of life science related subjects as well as basic medicine and clinical medical study, theoretically, it is more consistent with the ethics.
