Dynamic expression of CatSper1 in mouse testis during postnatal development and its relationship with human sperm motility

Abstract:Objective: To study the expression pattern of CatSper1 in postnatal mouse testis development and the relationship between the CatSper1 protein expression and human sperm motility.Methods: The temporal expression of CatSper1 mRNA in the mouse testis at day 11,15,18,21,25,28,35,42,56,and 120 postpartum(n=3 in each day group) were investigated using real time RT-PCR.Both high and low motile spermatozoa were isolated from the patients with normozoospermia(n=4) by four-layer Percoll gradients with the following densities: 95%,76%,57% and 47.5%,and then the expressions of CatSper1 protein in high and low motile spermatozoa were determined by Western Blot.Results: Very low levels of CatSper1 mRNA were detected in the mouse testes at day 18.Its expression increased sharply at day 25 and 28(2.95 and 7.59 fold as that at day 21 respectively),and increased gradually from day 42 onward,with the maximal expression at the adult stage.In every human semen sample,higher CatSper1 protein levels normalized by β-Actin in high motile sperm fractions than in low motile sperm fractions were observed. Conclusions: Significantly higher levels of CatSper1 protein in high motile sperm fractions than in low motile sperm fractions imply the role of CatSper1 in the sperm motility.The expression pattern of CatSper1 mRNA in postnatal mouse testis development will shed light on the interaction of the CatSper family members and their precise roles.
