Mechanism of G-CSF and AMD3100 enhancing the chemotherapeutic sensitivity of leukemia cells

Abstract:Objective: To investigate the effect and mechanism of G-CSF and AMD3100 on enhancing sensitivity of leukemia cells to chemotherapeutic drugs. Methods:HL-60 cells were pre-incubated at 37 °C for 48 h with G-CSF (50 ng/mL) and AMD3100 (5 μg/mL), alone or combined, and with medium only(as control). After washing, HL-60 cells were co-cul-tured with HS-5 supernatant or HS-5 cells. Cell viability was measured by Cell Counting Kit-8 (CCK-8). Apoptosis was as-sessed by flow cytometry using the Annexin V Apoptosis Kit. Cell surface CXCR4 and total CXCR4 protein were detected by flow cytometry and Western blotting, respectively. Real-time quantitative PCR was used to analyze the mRNA expres-sion. Results: HS-5 supernatant or HS-5 cells could protect HL-60 cells from spontaneous or drug-induced apoptosis. G-CSF alone and AMD3100 alone could partially inhibit such protective effect, and the combined use resulted in a syner-gistic effect. G-CSF could reduce the levels of cell surface CXCR4 protein, total CXCR4 protein and CXCR4 mRNA in cytoplasm, and up-regulated microRNA-146a. AMD3100 could only reduce surface CXCR4 expression, and total CXCR4, CXCR4 mRNA or microRNA-146a expressions were not affected. Conclusions: G-CSF and AMD3100 block the CXCR4/CXCL12 axis via down-regulating CXCR4 expression through different mechanisms, then disrupting the interaction be-tween bone marrow microenvironment and leukemia cells and sensitizing leukemia cells to chemotherapy.
