Initial study on apoptosis-inducing effect of baicalin on T-cell lymphoma

Abstract:Obejective: To investigate the effect of baicalin on T-cell lymphoma (TCL) and its underlying mecha-nism. Methods:Human TCL cell line Jurkat was incubated with baicalin. Cell growth inhibition was assessed by MTT as-say. Cell morphological changes were observed under microscopy with Wright's staining. Cell apoptosis was detected by flow cytometry. The expression levels of caspase-3, caspase-8, and caspase-9, and AKT/ERK-MYC cascade were evaluated by Western blot. The effect of combined baicalin with Doxorubicinwas was analyzed with Calcusyn software. Results:Baicalin inhibited Jurkat cell growth in a time- and dose-dependent manner, and its half inhibitory concentration at 48 h was (24.32±1.01) μg/mL. Baicalin-treated cells displayed significant apoptotic morphological changes. Flow cytometry also showed increased percentage of Annexin V+ cells after baicalin treatment, which could be converted by Pan-caspase in-hibitor ZVAD-FMK. With baicalin treatment for 48 h, expression levels of activated caspase-3, caspase-8, and caspase-9 in Jurkat cells were significantly up-regulated , while p-AKT, p-ERK, and MYC were down-regulated Furthermore, com-bined index plots showed synergistic interaction of baicalin with doxorubicin. Conclusions: Baicalin could efficiently in-hibit TCL cell growth, induce both intrinsic and extrinsic apoptosis, down-regulate AKT/ERK-MYC cascade, and synergis-tically interact with doxorubicin.
