Effect of Tumor Necrosis Factor-a on NF-kB Activity in Cultured Smooth Muscle Cells

Abstract:Objective To investigate the effect of tumor necrosis factor-a(TNF-a)on nuclear factor-k B(NF-Kb)activity in cultured vascular smooth muscle cells(SmGs).Methods Rat(n=12）aortic SMGs were cultured in DMEM containing 0.5% bovine caif serum for 48hours and then 25ng/ml of TNF-a was added into the medium for stimulation. Gells were colleced and nuclear protein of cells was extracted at 0, 15 and 30 minutes following stimulation. NF-kb activity in stimulated cells was examined by electrophoretic mobility shift assay (EMSA).Results Baseline NF-Kb nuclear binding in SMGs increased more than two-fold at 15 min(n=4,P<0.01)and three-fold at 30min(n=4,P<0.01)following stimulation of TNF-a.Conclusions TNF-a could enhance transient mobilization of NF-Kb in vascular SMGs,which suggested that some regulating effect of TNF-a on vascular SMGs might be exerted through NF-kB activation pathway.
