Abstract:Objective To study the effect of serum blood glucose (BG) and HbA1c levels on ischemic stroke. Methods Eighty ischemic stroke patients were divided into diabetes mellitus (DM) group 1 (n=58) and DM-free group 1 (n=22) according to their fasting blood glucose and DM history.Another 60 SD rats were divided into control group 2,DM-free group 2,DM group 2,pioglitazone treatment group and combined treatment group (12 in each group).Their tissue necrosis was observed with TTC staining, their scrum levels of BG,HbA1c,IL-6 and TNF-α were measured by ELISA.Expressions of PPARγ and NF-κB were detected by Western blot. Results The serum BG and HbA1c levels were significantly higher in DM group 1 than in DM-free group 1 and control group 1 (11.65±3.41 mmol/L vs 5.69±0.59 mmol/L and 4.23±0.52 mmol/L,9.13%± 2.21 % vs 5.57%±0.27% and 4.03% ±0.53%,P<0.05).The serum BG and HbA1c levels were significantly higher in DM-free group 1 than in control group 1.The size of ischemic stroke was significantly larger in DM-free group 2 and DM group 2 than in control group 2 (P<0.05) and in DM group 2 th.n in DM-free group 2 (P<0.05),and was significantly smaller in pioglitazone treatment group than in DM group 2 (P<0.05).Conclusion Elevated serum BG and HbA1c levels are closely related with ischemic stroke.Elevated serum BG level can aggravate ischemic stroke, which is related with the PPARγ-NF-κB signaling pathway.
