Abstract:Objective To explore the effect of the CD14 promoter gene-260 polymorphism in diabetic nephropathy(DN).Methods CD14 promoter gene C-260T polymorphism had been analyzed in four hunderd and thirty-seven T2DM patients and one hundred and forty-five normal controls by polymerase chain reaction direct sequencing.Results The distribution of CD14 promoter gene-260 polymorphsim and the allele frequency were no significant difference between T2DM group and normal control group(P0.05).The distributions of CC genotype and CT +TT genotype were revealed statistically significant difference in CD14/-260 between non-diabetic nephropathy group and diabetic nephropathy group(P0.05).Conclusions The distribution of CD14 promoter gene C-260T polymorphsim isn't associated with T2DM.The CC genotype of the C-260T polymorphism of CD14 gene is a genetic risk factor for diabetic nephropaty.
Relation of C-159T polymorphism in promoter region of CD14 and Korean patients with diabetic nephropathy in Yanbian area

Abstract:【Objective】 To investigate the distribution of CD14 promoter gene-159(C-T) polymorphism in Korean DN patients of Yanbian area,analyze the association of CD14 polymorphism with Korean DN patients in Yanbian area,and study the risk factors of DN.【Methods】 Analyzed the genotypes of CD14 in 152 Korean DN patients and 139 Korean of normal controls by polymerase chain reaction-restriction fragment length polymorphism,investigated the association between CD14 polymorphism and Korean DN patients in Yanbian area,and conducted a retrospective analysis of clinical features of 152 Korean DN patients.【Results】 ① The distribution of CD14 promoter gene-159(C-T) polymorphism showed no significant difference between normal control group and DN group(χ2=1.776,P >0.05).② The distribution of CC homozygous genotype vs.CT heterozygous and TT homozygous genotype showed statistically significant difference in CD14 C-159T between male and female Korean DN patients(χ2=3.903,P <0.05).③ DN had a close relation with hypertension,process,CRP fasting blood glucose and low density lipoprotein-cholesterol(P <0.05 or P <0.01),but had no relation with Age,PBG,GHb,Cr,CHO,TG,HDL-C and BMI(both P >0.05).【Conclusions】 ① The distribution of CD14 promoter gene-159(C-T) polymorphism has no relation with DN.② The CC genotype of the C-159T polymorphism of CD14 is a susceptibility gene for the male Korean DN patients.③ Hypertension,long process,high fasting blood glucose,low density lipoprotein-cholesterol ascension and inflammatory reaction are important risk factors for DN.
Relationship between CD14 C260T promoter polymorphism and serum C-reactive protein levels in patients with coronary artery disease

Abstract:Objective To investigate the relationship of serum levels of high sensitivity C-reactive protein(Hs-CRP) with CD14 C260T promoter polymorphism in patients with coronary artery disease(CAD) .Methods Serum levels of Hs-CRP were determined in 240 patients with coronary artery disease.C260T Promoter Polymorphism of the CD14 gene was screened by PCR-RELP assay.Results The genotype distribution of the C260T polymorphism of the CD14 gene was as follows:TT in 106 of 240 patients(44.2%) ,TC in 82 of 240 patients(34.1%) and CC in 52 of 240 patients(21.7%) .Patients with TT subtype had increased Hs-CRP levels compared with carriers of the C allele(P<0.05) .A higher percentage of T allele homozygotes had Hs-CRP levels >3 mg/L(P<0.05) .Conclusion This study indicated T allele homozygotes of CD14 gene may be an independent risk factor in systemic inflammation with stable CAD.
Association Between Polymorphism of C936T in Vascular Endothelial Growth Factor Gene and Type 2 Diabetic Nephropathy

Abstract:Objective To examine the genetic variations of the vascular endothelial growth factor(VEGF) gene and assess the possible relationship to diabetic nephropathy(DN) in type 2 diabetic mellitus (T2DM) patients in Chinese population. Methods The C936T polymorphism of VEGF gene were examined by polymerase chain reaction and restriction fragment length polymorphism in 296 Chinese subjects with T2DM,173 patients had DN and 123 were considered as control subjects (diabetic patients without DN). Results The genotype frequency of CT and TT was significantly higher in patients with DN than patients without DN (P 0.05). And the frequency of T allele in C936T polymorphism was also found to be significantly higher in patients with DN than patients without DN (P 0.01). Multiple logistic regression analysis revealed that TT and CT genotypes associated with diabetic nephropathy (OR=2.532, 95% CI=1.486 ～ 4.315,P=0.001). Moreover, present results suggested that CT and TT genotype had an effect on blood lipids given exposure to diabetic nephropathy. Conclusion The data suggest that the C936T polymorphism in 3'-untranslated region of VEGF could be one of the major genetic factors in increasing the risk for DN.

Abstract:OBJECTIVE:To investigate the distribution of CD14 promoter gene -159(C>T) polymorphism in Hubei Han population of China and analyze the association of CD14 polymorphisms with coronary heart disease (CHD). METHODS:Genotypes of CD14 were typed in 162 CHD patients and 196 controls by polymerase chain reaction-restriction fragment length polymorphism. Selected coronary angiography was performed in 162 CHD patients. RESULTS:CD14 promoter -159 genotype frequencies of CC, CT and TT were 27.4%, 45.6%, 27.0% and 14.8%, 46.5%, 38.7% in normal control group and CHD group respectively. Genotype distribution was in accordance with Hardy-Weinberg equilibrium. There existed statistically significant difference in frequencies of allele and genotype in CD14 C-159T polymorphism between CHD group and control group (Genotype: Chi2=0.654, P < 0.05, CT vs CC, OR=1.245, 95%CI: 1.001-1.473, TT vs CC, OR=2.374, 95%CI 2.012-2.649; Allele: Chi2=0.547, P < 0.05, T vs C, Chi2=0.547, P < 0.05, OR=3.105, 95%CI: 2.493-3.539). The distributions of allele and genotype in CD14 -159(C>T) were of statistically significant difference between non-myocardial infarction subgroup and myocardial infarction subgroup (Genotype: Chi2=0.782, P < 0.05, CT vs CC, OR=2.375, 95%CI: 2.017-2.689, TT vs CC, OR=3.459, 95%CI: 3.003-3.846. Allele: Chi2=2.374, P < 0.05, T vs C, Chi2=2.374, P < 0.05, OR=4.011, 95%CI: 3.814-4.279). However, no statistically significant difference was found among the subgroups of oneìtwo and three stenosed vessels. CONCLUSION:The T allele of the C-159T polymorphism of CD14 gene may be a risk factor for myocardial infarction.

Abstract:AIM: To investigate the association between the C(-159)T polymorphism in the promoter of CD14 gene and coronary heart disease (CHD) in the Han population of Jiangsu province. METHODS: Between August 2003 and September 2005, 193 CHD patients were recruited from Nanjing Drumtower Hospital. They all had > 50% stenosis in at least one major coronary artery and were diagnosed by coronary angiography. Meanwhile 225 uncorrelated subjects who received routine health examination in the same hospital were taken as healthy control group. Using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method, the C(-159)T polymorphism in the promoter of CD14 gene were analyzed in CHD and control groups, the levels of serum lipid and soluble CD14 were also measured by biochemical and enzyme linked immunosorbent assay methods respectively. RESULTS: All the subjects completed the test and entered the result analysis.①The levels of serum lipids and soluble CD14: Compared with control group, CHD group had higher levels of serum total cholesterol, triglyceride, low-density lipoprotein cholesterol, and CD14, but the level of high-density lipoprotein cholesterol was lower in CHD group.②Genotype analysis: 3 genotypes were observed in the study: CC genotype, CT genotype, and TT genotype were 20.90%, 55.10%, 24.00% and 15.00%, 49.20%, 35.80% in control group and CHD group, respectively. And the control group was significantly higher than CHD group (χ2 =7.527, P < 0.05).③The genotype frequencies of C and T alleles were 48.40%, 51.60% and 39.40%, 60.60% in control group and CHD group, respectively. The difference between the two had a statistical significance (χ2 =6.858, P < 0.05).④No significant differences for the levels of serum lipids and soluble CD14 were found among TT, TC, and CC genotypes in CHD patients (P > 0.05). CONCLUSION: The C(-159)T polymorphism in the promoter of CD14 gene is associated with CHD in the Han population of Jiangsu province, and it might be a genetic predisposing predictor for CHD in the Han population.

Abstract:Objective To examine whether C(-260)T polymorphism in the promoter of CD14 gene is associated with coronary heart disease(CHD)in the Han population of Jiangsu region.Methods Applying polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)methods we analyzed the genotypes of 193 CHD patients,whose coronary lesions were confirmed by angiography,and 225 healthy subjects.The levels of serum lipid and sCD14 were studied by biochemical methods and ELISA method respectively.Results The frequencies of TT,CT and CC genotype of CD14 gene promoter-260 polymorphism were 41.78%,40.89%,17.33% in healthy subjects(control group)and 40.41%,49.23%,10.36% in CHD group,respectively.No significant association was noted in the distributions of genotype and the frequencies of the allele in the site between CHD and control group(P>0.05).Conclusions The C(-260)T polymorphism in the promoter of CD14 gene was not associated with CHD in the Han population of Jiangsu region,and it might not be a genetic risk factor of CHD in the Han Chinese.

Abstract:Objective To evaluate the role of RANTES receptor(CCR5) gene promoter polymorphism on diabetic nephropathy in Chinese type 2 diabetic nephropathy patients.Methods Type 2 diabetes mellitus patients and diabetic nephropathy(including diabetes mellitus with microalbuminuria and diabetes mellitus with macroalbuminuria) and controls were recruited.CCR5 59029 gene were detected by PCR-restriction fragment length polymorphism(PCR-RFLP) method,The association of genotypes with diabetic nephropathy was analyzed.Results The frequency of a CCR5 59029 positive genotype(G/A or A/A) was higher in the DN2 group than in the DNO group or DN1 group(P=0 and P=0.02) and the frequency of CCR5 59029 positive genetype was higher in the DN(DN1+DN2) than in the DNO group(P=0.007).Conclusion The CCR559029 may be an independent risk factor for diabetic nephropathy in patients with type 2 diabetes and may have an additive effect on nephropathy.

Abstract:OBJECTIVE:To investigate whether the angiotensin II type I receptor gene (AGTR1) A1166C polymorphism is associated with a high risk of diabetic nephropathy.METHODS:The allele frequency and the genotype distribution of the AGTR1 A1166C polymorphism were studied in normal controls (157 cases), simple diabetes (141 cases, duration of diabetes >10 years), and diabetic nephropathy (152 cases) by means of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).RESULTS:Patients with diabetic nephropathy had a higher frequency of C allele of the AGTR1 A1166C polymorphism than that of normal controls and simple diabetes (P<0.05); but there was no significant difference in frequency of C allele between the normal controls and patients with simple diabetes.CONCLUSION:The diabetic patients with AGTR1 C allele may be more susceptible to diabetic nephropathy than diabetic patients with A allele.
The Role of CD14 159C/T Polymorphism in Susceptibility of Cancers

Abstract:CD14 (cluster of differentiation 14), a kind of lipopolysaccharide (LPS) receptor, plays an important role in the innate immune system. Up to now, accumulating studies have reported the association between CD14-159C/T polymorphism and susceptibility of cancers, with divergent results. We thus conducted this meta-analysis to demonstrate the possible association between CD14-159C/T polymorphism and cancer risk. All eligible case control studies were identified by searching PubMed, Embase and CNKI database up to November 2017. Pooled odds ratio (OR) was used to access the strength of association. Subgroup analysis was also conducted based on cancer type, ethnicity and language. Statistical analyses were performed by using STATA 12.0 software. A total of 21 case-control studies involving 11241 participants (4136 cases and 7105 controls) were included in the current meta-analysis. The result indicated that the T allele of CD14-159C/T polymorphism did not confer risk for overall cancers (TT + CT vs. CC: OR = 0.98, 95% CI = 0.83-1.16, P = 0.81; TT vs. CT + CC: OR = 1.13, 95% CI = 0.87-1.46, P = 0.37). However, a subgroup analysis by cancer type indicated the CD14-159C/T polymorphism was significantly correlated with acute lymphoblastic leukemia (TT vs. CT + CC: OR = 1.80, 95% CI = 1.37-2.38, P < 0.001; C vs. T: OR = 0.81, 95% CI = 0.67-0.98, P = 0.03) and prostate cancer (TT vs. CC: OR = 0.62, 95% CI = 0.39-0.99, P = 0.04). Nevertheless, we failed to detect any relationship in the subgroup analysis by ethnicity and language. This meta-analysis suggested that CD14-159C/T polymorphism may be associated with acute lymphoblastic leukemia and prostate cancer, but no significant relationship was found between CD14-159C/T polymorphism and overall cancer. Further well-designed studies with lager sample size on different cancer types and ethnicities are needed.

Abstract:Although the role of CD14 in recognizing Mycobacterium tuberculosis is well-understood, the possible role of polymorphisms in susceptibility to develop tuberculosis remains unclear. This study evaluates whether there is an association of polymorphisms within the promoter of the CD14 gene with susceptibility to pulmonary tuberculosis. In a casecontrol study, we genotyped the eight known single nucleotide polymorphisms SNPs within the promoter of the CD14 gene of 698 Han Chinese subjects. Statistically significant differences between tuberculosis patients and healthy controls were found for G-1619A, T-1359G, A-1145G, and C-159T. The haplotype-GGGT, composed of these four SNPs, exhibited a significant association with the disease. Furthermore, expression levels of soluble CD14 were significantly higher in tuberculosis patients with the GGGT haplotype than with other haplotypes, while IgE expression levels were significantly reduced. Our results suggest that these four SNPs within the promoter of the CD14 gene are associated with susceptibility to pulmonary tuberculosis.
Association of CD14 Gene Polymorphism with Atopic Diseases in Chinese Han Ethnic Group Children

Abstract:OBJECTIVE:To investigate the distribution characteristics of the single nucleotide polymorphisms (SNPs) of the human CD14 gene in Chinese Han ethnic group children in Wenzhou, and their association with atopic diseases.METHODS:Totally 113 cases were recruited in atopic disease group who met the following criteria: 2 - 12 years old, clinically diagnosed as asthma or allergic rhinitis or atopic dermatitis, elevation of serum total IgE levels and serum specific IgE. Sixty-seven healthy children were enrolled in control group. The related regions of CD14 gene were sequenced to identify and characterize the SNPs, and plasma TIgE and SIgE were detected by immunoassay system and uniCAP system, respectively. The frequency of genotypes and alleles between two groups, as well as the levels of IgE in different genotypes, were compared.RESULTS:CD14/-159 SNP was present in Han ethnic group population of Wenzhou. The frequency of each genotype was 57.0% (TT), 28.0% (TC), 15.0% (CC) in normal children, and 46.9% (TT), 35.4% (TC), 17.7% (CC) in atopic children. No significant difference was found in the distribution of CD14/-159 polymorphism between atopic children and healthy control (chi(2) = 1.918, P > 0.05) according to Hardy-Weinberg principle statistics. There were no significant difference in frequency of each genotype between boys and girls. No significant difference was found in the total plasma IgE levels among groups of TT genotypes [(2520 +/- 460) IU/L], TC genotypes [(2400 +/- 460) IU/L] and CC genotype [(2500 +/- 460) IU/L] (F = 0.807, P > 0.05).CONCLUSION:CD14/-159 SNP is present in Han ethnic group children in Wenzhou, and other SNP in CD14 gene was not found. TT genotype was the primary genotype in CD14/-159 SNP in the children studied. No relationship between CD14/-159 SNP and atopic disease or serum total IgE level was found.

CD14 Genomic Polymorphism Influences CD14 Expression in Whole Blood Culture

Abstract:OBJECTIVE:To investigate whether -159C/T promoter polymorphism of the CD14 gene influences the CD14 expression as well as release in whole blood culture.METHODS:One hundred and eighteen healthy human blood donors were included in the present study. CD14 mRNA expression and soluble CD14 levels were measured using a whole blood cell culture model with or without lipopolysaccharide (LPS) stimulation. The CD14 gene polymorphism was determined by polymerase chain reaction and subsequent HaeIII restriction enzyme digestion of the polymerase chain reaction products. TNF-alpha production in the whole blood culture was also measured.RESULTS:Among the 118 individuals, there were 40 subjects homozygous for the T allele (TT), 62 were heterozygous (CT), and 16 had the genotype CC. The CD14 mRNA expression in leukocytes and soluble CD14 levels in supernatant were higher in TT homozygotes and carriers of the genotype TC compared with individuals homozygous for the C allele (P < 0.05 or 0.01). In addition, the highest values of TNF-alpha production were found in TT homozygote [(352 +/- 215) pg/ml] compared with both genotype TC and CC [(261 +/- 163) pg/ml, (198 +/- 122) pg/ml, both P < 0.05].CONCLUSION:The -159C/T promoter polymorphism of the LPS receptor CD14 may influence the CD14 expression as well as release in whole blood culture, and it might be associated with TNF-alpha response to LPS stimulation.
EFFECTS OF CD14-159 C/T POLYMORPHISM ON CD14 EXPRESSION AND THE BALANCE BETWEEN PROINFLAMMATORY AND ANTI-INFLAMMATORY CYTOKINES IN WHOLE BLOOD CULTURE

Abstract:CD14 is an important receptor of innate immunity. When CD14 is anchored by ligands to LPS, peptidoglycans, or lipoteichoic acid, it can result in either proinflammatory or anti-inflammatory responses. To determine whether CD14-159 C/T polymorphism is associated with CD14 expression and the balance of proinflammatory and anti-inflammatory responses, we studied 118 healthy ethnic Han Chinese using a whole blood culture model. The CD14-159 C/T polymorphism was determined by polymerase chain reaction and subsequent HaeIII restriction enzyme digestion of the polymerase chain reaction products. Meanwhile, CD14 mRNA expression in leukocytes and the levels of soluble CD14, TNF-alpha, IL-6, and IL-10 were also determined in the supernatants. Among the 118 individuals, there were 40 TT homozygotes, 62 heterozygotes, and 16 subjects homozygous for C allele. After LPS stimulation, the levels of CD14 mRNA expression in TT and TC genotypes were significantly higher than in CC homozygotes (P = 0.017), and soluble CD14 levels were also higher than in CC genotypes (P = 0.008). In addition, TT homozygotes had the highest LPS-stimulated TNF-alpha, IL-6 production (P = 0.044, P = 0.004), and the lowest IL-10 release (P = 0.003). In conclusion, CD14-159 C/T polymorphism is correlated with CD14 expression and may thus influence the balance of proinflammatory and anti-inflammatory responses in ethnic Han Chinese. These results suggest that CD14-159 C/T polymorphism might partly explain the difference in predisposition to develop complications of infectious diseases in different patients and may provide a therapeutic target for sepsis intervention strategies.
Relationship Between Uncoupling Protein 2 Gene Promoter-866 G/A Polymorphism and Chinese Diabetic Nephropathy

Abstract:Objective To investigate the relationship between uncoupling protein 2 (UCP-2) gene promoter -866 G/A polymorphism and Chinese diabetic nephropathy (DN). Methods A total of 182 patients with type 2 diabetic mellitus were selected and divided into DN group (90 cases with DN ) and NCD group (92 cases without DN ). Genomic DNA was detected, and UCP-2 genotype and allele gene frequency was confirmed by polymerase chain reaction-restrictive fragment length polymorphism, then compared.Results The genotype frequency of UCP-2 gene promoter-866 G/A polymorphism was distributed in DN group[GG 14.44%( 13/90),GA 31.11%(28/90),AA 54.44%(49/90)],and distributed in NCD group[GG 29.35% ( 27/92 ), GA 32.61% ( 30/92 ), AA 38.04% ( 35/92 )], and there was significant difference between two groups ( χ2 = 7.28 , P ＜ 0.05 ). There was also significant difference in allele gene frequency between DN group and NCD group[G 45.65% (84/184) vs. 30.00% (54/180),A 54.35% (100/184) vs. 70.00% (126/180)]( χ2 = 9.47, P ＜ 0.05 ). Conclusion There is correlation between the UCP-2 gene promoter-866 G/A polymorphism and Chinese DN, and the incidence of DN with A/A genotype is increased.
Association Between Cluster of Differentiation 14 Polymorphism and Susceptibility of Infection

Abstract:OBJECTIVE:To investigate the association between the single nucleotide polymorphisms (SNPs) of cluster of differentiation 14 (CD14) and susceptibility in patients with sepsis.METHODS:One hundred and twelve patients with sepsis admitted to the intensive care unit (ICU) from December 2004 to March 2006 were enrolled for study. Another 100 healthy volunteer donors were chosen serving as the normal control group. Genotypes and alleles of CD14-159 C/T, CD14-260 C/T were determined in patients and healthy controls by means of restriction fragment length polymorphism (RFLP) analysis of polymerase chain reaction (PCR) products. Distributional differences of genotypes and alleles between patients and healthy controls were analyzed. The sepsis patients were divided into survivor group and non-survivor group depending on survival for 28 days. Distributional differences of genotypes and alleles between survivor group and non-survivor group were determined.RESULTS:The rates of CD14-159 C/T genotypes and T-alleles in patients with sepsis were significantly higher than those of healthy volunteers (P=0.005 and P=0.000). There were no significant differences in the rates of CD14-260 C/T genotypes and T-alleles in two groups (P=0.912 and P=0.706). The rates of CD14 -159 C/T genotypes and T-alleles in non-survivors were significantly higher than those of survivors (both P=0.000). There were no significant differences in the rates of CD14-260 C/T genotypes and T-alleles in two groups (P=0.643 and P=0.890).CONCLUSION:The differences of CD14-159 C/T genotypes of SNPs may be associated with the higher risk of susceptibility and mortality of sepsis.

Abstract:Background: The -159C/T polymorphism in the CD14 gene has been implicated in susceptibility to tuberculosis, but the results were inconclusive. The present meta-analysis aimed to perform a comprehensive assessment of the literature on the possible association between the -159C/T polymorphism and tuberculosis risk.Methods: We searched in Pubmed and Embase for studies evaluating the association between the -159C/T gene polymorphism and tuberculosis risk. Data were extracted and statistical analysis was performed using Revman 5.1 and STATA 12.0 software.Results: A total of seven case-control studies involving 3253 subjects (1,574 tuberculosis cases and 1,679 controls) were included. Combined analysis revealed an obvious association between this polymorphism and tuberculosis risk (odds ratio=1.66 and 95% confidence interval: 1.23-2.25, P<0.05 for TT vs. TC+ CC). Sub-group analysis by ethnicity suggested that the risk of tuberculosis associated with the -159C/T polymorphism was significantly elevated among Asians (odds ratio=1.87 and 95% confidence interval: 1.58-2.21, P<0.05 for TT vs. TC+ CC).Conclusion: This meta-analysis suggests that the -159C/T polymorphism in the CD14 gene contributes to tuberculosis susceptibility. To further investigate gene-gene and gene-environment interactions between this polymorphism and tuberculosis risk, more studies are needed.
Polymorphism of CD14 Gene but Not the Mutation of TLR4 Gene is Associated with Colorectal Cancer in Chinese Patients

Abstract:Objectives: Toll-like receptor 4 and CD14 are components of the lipopolysaccharide receptor complex. Our study aimed to investigate an association between TLR4 Asp299Gly and CD14-260 polymorphisms in Chinese patients with colorectal cancer.Method: By a method of polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP), we genotyped TLR4 Asp299Gly and CD14-260 polymorphisms in 110 unrelated patients with colorectal cancer and 160 healthy controls from the Chinese Han population.Results: We found significant differences in CD14 genotypes between healthy controls and patients with colorectal cancer. The frequency of the C/C genotype in healthy controls (15.6%) was significantly lower than in the group of colorectal cancer patients (32%). The frequency of the C/T genotype in healthy controls (48.1%) was significantly higher than that in the group of colorectal cancer patients (31%). No TLR4 Asp299Gly mutation was detected in any patients or healthy controls in the Chinese Han population.Conclusions: These findings indicated that the polymorphism of CD14 but not TLR4 Asp299Gly mutation was associated with Chinese patients with colorectal cancer, and the CD14 gene may contribute to the predisposition to colorectal cancer. Screening for the CD14 C-260T genotype is likely to be a useful tool for risk assessment or prognostication for colorectal cancer in the Chinese Han population.
Association Of The Transcription Factor 7 Like 2 (Tcf7l2) Polymorphism With Diabetic Nephropathy Risk A Meta-Analysis

Abstract:It is assumed that genetic factors may participate in the development of diabetic nephropathy (DN). The association between TCF7L2 gene polymorphism and DN risk is still unclear. To evaluate the relationship, we performed this meta-analysis.Eligible relevant studies were searched and selected from PubMed, Embase, and ISI Web of Science. Summary effect estimates were derived using a random effects model, with attention to study quality and publication bias. Ethnical approval was not necessary, because this meta-analysis was based on published articles, and did not involve patient consent.A total of 7 studies were identified. Analysis of all studies indicated significant association between TCF7L2 gene polymorphism and DN risk (odds ratio [OR] = 1.31, 95% confidence interval (CI) = 1.10-1.56, P-heterogeneity < 0.00001, P = 0.002). Subgroup analysis showed similar results in Asian (OR = 1.33, 95% CI = 1.10-1.62, P-heterogeneity = 0.03, P = 0.004), in Caucasian (OR = 2.27, 95% CI = 1.78-2.90, P-heterogeneity = 0.17, P < 0.00001), in rs7903146 mutation (OR = 1.61, 95% CI + 1.25-2.07, P-heterogeneity < 0.00001, P = 0.0002), However, no association was observed in Negroid (OR = 1.10, 95% CI = 0.90-1.35, P-heterogeneity< 00001, P = 0.36).Our results suggest that TCF7L2 gene polymorphism may contribute to the risk of DN. However, more studies should be launched in the future.
