Abstract:Objective To investigate the effect of seal oil on galectin-3 in serum and liver in rats with nonalcoholic steatohepatitis(NASH). Methods 24 male SD rats were randomly divided into three groups: normal group fed with standard diet, model group and treated group fed with high-fat diet consisting of 88 g standard diet,10 g lard oil,and 2 g cholesterol. The treated group was given seal oil(1.0 g/d) by intragastric administration after 8 weeks. After 12 weeks, all rats were killed.Body weight, body length, and liver weight were recorded. The contents of TNF-α and galectin-3 in serum and liver were detected with ELISA.The expression of galectin-3 mRNA in liver was evaluated by real-time PCR.Hepatic histology was examined by HE staining. Results The model rats developed obesity,NASH with hepatomegaly, and a significantly higher concentrations of galectin-3 in serum and liver, serum TNF-α content and galectin-3 mRNA expression.Compared with the model group, the treated group showed decrease of the indexes, statisticaly. In addition, the prevalence of NASH in treated group was significantly lower than that in model group(87.5% vs 100%,P0.01).Pearson correlation analysis revealed a positive correlation between liver galectin-3 contents and liver weight(r=0.881,P0.01). Conclusion Galectin-3 is associated with the development of obesity related hepatic lipid deposition.Seal oil can significantly decrease the expression of galectin-3 of NASH rats, which may be one of its vital therapeutic mechanisms.
