Nuclear Transcription Factor Foxo1 and Glucose,lipid Metabolism

Abstract:Forkhead box 01 (Foxo1) is a nuclear transcription factor that be longs to a protein family characterized by a highly conserved DNA binding motif, termed the forkhead box. Foxo1 is a substrate of phosphatidylinositol 3-hydroxy kinase/protein kinase B and plays an negative role in insulin signaling pathway. Animal test and clinical research show that the expression of Foxo1 is significantly increased in hepato-cyte nuclear in insulin resistant and fatty liver states. While accompanying with the improvement of insulin resistance and weight reduction,the expression of Foxo1 tends to normal,and then Foxo1 transports out of cell and lose activity. Studies also show that inhibition of the expression of Foxo1 can significantly improve the blood glucose,dyslipidemia and the accumulation of triglyceride in liver. It indicates that controlling the expression of Foxo1 may be a target for treatment of diabetes,dyslipidemia and insulin resistance.
