Experimental study of the metal-regulatory transcription factor-1 on osteoclast differentiation and function

Abstract:Objective:To study the effect of metal-regulated transcription factor-1 (MTF-1) on osteoclast differentiation and function and explore its possible molecular mechanism.Methods:Quantitative polymerase chain reaction (qPCR) was used to detect changes in MTF-1 expression during osteoclast differentiation. Small molecule inhibitor LOR-253 was applied to inhibit the transcriptional activity of MTF-1, tartrate-resistant acid phosphatase (TRAP) staining, qPCR, bone plate resorption test was used to detect changes in osteoclast differentiation and function. ANOVA and SNK test were used to analyze the differences between groups.Results:One day after the induction of receptor activator of nuclear factor-κB ligand (RANKL), the expression of MTF-1 in RAW264.7cellsincreased by 5.13 times. After LOR-253 down-regulated the transcriptional activity of MTF-1, the number of osteoclasts was significantly reduced (

P<0.05), with (74.6±3.2) cells in the control group, (35.2±1.7) cells in the 50 nmol/L group, (9.2±0.7) cells in the 100 nmol/L group; TRAP enzyme activity was also significantly reduced (

P<0.05); bone plate resorption area was significantly reduced (

P<0.05), the control group was (75.0±6.3)%, the 50 nmol/L group was (49.7±3.4)%, the 100 nmol/L group was (15.9±1.4)%; the expression of osteoclast marker genes, TRAP, cathepsin K (CK), matrix metalloproteinase-9 (MMP-9) and nuclear factor of activated T cells-1 (NFATC1), werealso down-regulated. In the 50 nmol/L group: 0.70±0.02, 0.65±0.02, 0.34±0.01, 0.80±0.01; in the 100 nmol/L group: 0.60±0.01, 0.40±0.02, 0.30±0.01, 0.75±0.01 (

Conclusion:Our study found that the inhibition of MTF-1 transcriptional activity by LOR-253 significantly inhibited osteoclast differentiation and function, and NFATC1 played an important role in it.
