Effect of Infection with Toxoplasma Gondii on the HLA-G Expression in the Early Pregnant Human Trophoblastic Cells

Abstract:Objective To investigate the effect of infection with Toxoplasma gondii on the human leukocyte antigen G(HLA-G) expression in the human trophoblastic cells and to explore the immune mechanisms in abnormal pregnancy induced by T. gondii. Methods The trophoblastic cells were isolated from the villous tissues, which were obtained from induced abortions of normal pregnancies for 6 to 12 weeks. The trophoblastic cells were divided into experimental and control groups. The experimental group was infected with T. gondii(1 × 104), and the control group was without infection. After being incubated for 48 h, the expression of HLA-G mRNA was analyzed with quantitative real-time PCR. Results Experimental results showed that the average expression levels of HLA-G mRNA in trophoblastic cells were 0. 15 +0.04 in the experimental group,while those in the control group were 0. 29 ± 0. 07. On protein level, the average ratio of HLA-G positive subgroup in the experimental and control group were (12.19 ± 2.04) % and (24.24 ± 3.07) %, respectively.The differences in these groups were statistically significant (P < 0.01). Conclusion From the above observations, it is evident that during the early stage of pregnancy, the expression of HLA-G in the trophoblastic cells was decreased in both mRNA and protein levels due to the infection of T. gondii.
