Influence of Alendronate on Proliferation of Rat Steoblasts

Abstract:Objective: To study the applicability of Alendronate for the prevention of the prosthesis aseptic loosening by investigating the effect of alendronate on osteoblasts.Method: The second generation of rat osteoblast wase cultured in medium with different concentration( 1 × l0~-10 mol/L,1×10~-9 mol/L,1×10~-8 mol/L,1×10~-7 mol/L,1×10~-6 mol/L,1×10~-5 mol/L,1×10~-4 mol/L) of Alendronate .Cell proliferation was detected by MTT method.Result: Alendronate inhibited proliferation of osteoblast at concentation of 10~-4 mol/L and 1×10~-5 mol/L,had no effect on proliferation of osteoblast at concentation of concentration of 10~-9 mol/L and l0~-10 mol/L,and increased osteoblastic proliferation at concentration of 10~-6 mol/L,10~-7 mol/L,10~-8 mol/L ,Peak levels were gained at concentration of 10~-8 mol/L.Conclusion:Alendronate has an anabolic effect on human osteoblast.Moreover,the effect relies on a proper concentration of Alendronate.
