The Effect of Culture in Vitro on the Biological Behavior of Human Bone Marrow Mesenchymal Stem Cells

Abstract:Objective To explore the biological behavior of human bone marrow mesenchymal stem cells(MSCs) after cultured in vitro. Methods MSCs from 5 persons were cultured and subcultured in vitro. The 2nd, 3rd and 9th subcultured cells were observed morphologically, and detected for cycle parameters of proliferation with flow-cytometer, respectively. And, the first to 6 th subcultured cells were tested with concanavalin A(Con A). Another 3rd and 9 th cells were inoculated in severe combined immune deficiency mice subcutaneously. At 1,2,3,4 week after inoculation, the mice were killed,and the tissues of inoculated site were examined with microscope. Results After long term culture in vitro,the cells shape obviously changed,and the hallmarks of senescence appeared in the cell. A lot of cells were heaped at the DNA synthesis phase. But, the cells were still diploid. No agglutination could be found, when the cells of the ninth passage were contacted with the ConA. After inoculated, the MSCs could produce collagen without inflammation reaction. The collagen produced by the third passage cells were more compact than that produced by the ninth passage cells. The cells were localized, and did not infiltrate into the surrounding tissue. 4 weeks later, the cells and the collagen they formed disappeared.Conclusions After long period culture in vitro,although the ability of proliferation of MSCs decrease,the cells do not acquire the tumor-like characteristics. The cells can be re-implanted into human body safely.
