Establishment of Syngeneic 9l/fischer344 Rat Brain Glioma Model

Abstract:Objective To establish the syngeneic rat brain glioma model of 9 L/Fischer344. Methods The 9L gliosarcoma cells were injected stereotactically into the right caudate nuclear of syngeneic Fischer 344 rat. Thirty-five rats were divided into 3 groups in random. Each rat received 5.0×10 2 , 1.0×10 5 and 1.0×10 6 9L cells in a volume of 10~20 μl, respectively. Implantation was followed by general observation. The survival time of each rat was recorded . MRI scanned on 10, 14 and 20 days after implantation for two rats received 1.0×10 5 9L cells. After the natural death, rat brains were sectioned and inspected. The tumor-containing samples were prepared by haematoxylin -eosin(HE), immunohistochemiscal glial fibrillary acidic protein (GFAP) and S-100 protein stains. Tumor pathological characteristics were investigated with microscope.Results ①All rats died within 45 days after injection of 9L cells. The median survival times of 3 groups were 38, 24 and 15 days, respectively. ②At 10, 14 and 20 days after implantation, brain tumors were detected clearly by MRI with gadolinium diethylene triaminepentaacetic acid (Gd-DTPA). The tumor sizes increased constantly during the observation. ③Demarcated massive tumors were found in the brains of all rats without extracerebral metastasis and extracranial extension. Central hemorrhage and necrosis was commonly seen in samples. Morphology and microscopic structures for glioma, such as no encapsulation, intraparenchymal growth, neovascularity, were observed in HE stain sections. Immunohistochemistory GFAP and S-100 protein stains of tumor cells were negative. Conclusion The syngeneic 9L/Fischer 344 rat brain glioma model is successfully established. After implantation, globular brain-tumor growth occurs constantly without extracranial extension . The tumor pathological characteristics mimic human malignant glioma.
