HEPATIC ALLOANTIGENIC UPTAKE IN PORTAL VENOUS IMMUNOTOLERANCE IN BALB/C RATS

Abstract:OBJECTIVE To explore the role of hepatic uptake of alloantigen in the mechanisms of immunotolerance induced by portalvenous administration. METHODOLOGY Spleen cells(107/L) depleted of erythrocytes from NIH/q mice were used as a tol-erated antigen in BALB/C rats by administration through portal vein. Gadolinium chloride(GdCl3,7mg/kg body weight), known forits ability to inhibit Kupffer cell phagocytosis, was given by portal vein to prevent the induced tolerance. Seven days after the induc-tion, BALB/C rots were inoculated on the dorsal flanks with erythrocytes free spleen cells(107/L). Mixed lymphocyte reaction(MLR)of BALB/C rots to NIH/q mice alloantigens was investigated, and delayed-type hypersensitivity(DTH) response to NIH/qmice alloantigens was assessed by direct challenging BALB/C rots footpad 7 days later with NIH/q spleen cells(107/L) free of ery-throcvtes. RESULTS Both systemic venously injected neuraminidase-treated allogenic spleen cells,and portal venously injecteduntreated allogenic spleen cells, significantly abolished BALB/C rats'capacity to develope MLR and DTH response to NIH/q mice's alloantigen. With simutaneouly injected GdC13 which inhibited Kupffer cell's phagocytosis, neither systemic venously injected neu-raminidasetreated allogenic spleen cells, nor portal venously injected untreated allogenic spleen cells, significantly abolishedBALB/C rots'capacity to develope MLR and DTH response to NIH/q mice's alloantigen. CONCLUSION Selective alloanti-gen uptake by Kupffer cells in the liver of BALB/C rots is essential for the induction or prevention of portal venous tolerance toNIH/q rat's spleen cells.
