Abstract:Background: Clinical observation suggested that micro-negative pressure drainage dressing (MNPDD) can promote refractory wound healing. This study explored the effect of MNPDD on refractory wound.Methods: Human keratinocytes (HaCat) and human fibroblasts (HF) were used to evaluate cell migration ability with micro-negative pressure. MNPDD promotion of cell migration in vivo was reflected by the vimentin content in rat. A multi-centre retrospective cohort study evaluated the effect of MNPDD by comparing skin grafting rate.Findings: Cell experiments revealed that with micro-negative pressure, HaCat and HF had higher migration capacity at 24 h (22·10±4·24% vs 1·26±1·65% for HaCat and 26·08±4·30% vs 1·98±1·82% for HF, P<·001). Animal experiments revealed that the wounds with MNPDD had higher vimentin concentration (43·8±4·6 vs 22·5±4·0 per field of view at day 9, P<·001).
