Abstract:OBJECTIVE The over expression of hepatitis BX-interacting protein(HBXIP) is detected in a variety of tumors and it is closely related with the development,invasion and metastasis of tumors.The purpose of this article was to explore the correlation between the over expression of HBXIP and the tumor size,lymph node metastasis,clinical stage,pathological grades and prognosis of bladder urothelial carcinoma (BUC).METHODS The cases came from Yanbian University Hospital with complete clinical and pathological information from January 1,2010 to December 31,2014.Totally 100 cases of BUC tissue were as a study group,10 cases of adjacent normal tissue were as a control group in the same period.Another 10 cases of frozen BUC tissue and matched paracarcinoma tissue were collected as study group and control group.Immunohistochemical SP method,Western blot and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of HBXIP.The correlations between the HBXIP expression and the clinical pathological features and prognosis of the patients were analyzed.RESULTS The immunochemistry results showed that the over expression of HBXIP was 86.0％ (86/100) in BUC and 20.0％ (2/10) in adjacent normal tissues respectively with a significant difference between two groups (P=0.036).The over expression of HBXIP was 79.3％ (23/29) in muscle invasive group and 50.7％ (36/71) in non-muscle invasive group,the difference was significant (P=0.046).Western blot showed that the relative quantitative expression of HBXIP was 0.712±0.135 in BUC and 0.326±0.075 in paracarcinoma tissue,the difference was significant (P =0.047).RT-PCR showed that the quantitative expression of HBXIP mRNA was 5.46±0.34 in BUC and 1.01±0.20 in paracarcinoma tissue,the difference was significant (P=0.041).The over expression of HBXIP was positive correlation with the depth of tumor invasion (P=0.048) and lymph node metastasis (P 0.036).We also found that the postoperative recurrence rate was 42.5 ％ (20/47) in over expression group and 15.1 ％ (8/53) in low expression group with a significant difference between two groups (P=0.003).Univariate analysis showed that overall survival rate was significantly lower in BUC patients with high level HBXIP expression than those with low level of HBXIP expression (P=0.014).The survival rate was significantly lower in T2-T3 stage than those in T0-T1 stage (P=0.023).Multivariate analysis was done by Cox proportional hazards model and it showed that the expression of HBXIP might be the independent risk factor of the prognosis of BUC (P=0.031).CONCLUSIONS Over expression of HBXIP was verified in BUC,which was closely related to the development,invasion and metastasis of BUC.Over expression status of HBXIP could detect the poor prognosis of BUC.
