The protective effect of tanshinone-I on radiation-induced bladder injury

Abstract:Objective To investigate the protective effect of tanshinone-I (T-I) on radiation-induced bladder injury (RIBI).Methods In vivo study:The female ICR mice were divided into 4 groups:control group 1 (control),control group 2 (T-Ⅰ),model group 1 (R),and model group 2 (R+T-Ⅰ).Mice in the T-Ⅰ and R+T-Ⅰ groups received intraperitoneal injection of T-Ⅰ,while mice in the control group and R group received olive oil.After 2 weeks,the mice were exposed to radiation.Urinary bladder samples were collected after 1 week or 12 weeks.Superoxideismutase (SOD),malondialdehyde (MDA)and bladder histopathology score were compared among the groups.In vitro study:the human uretherial cell line (SV-huc-1)was divided into 4 groups:control group 1 (control),control group 2 (T-Ⅰ),model group 1 (R) and model group 2 (R+T-Ⅰ).Group T-Ⅰ and R+T-Ⅰ were treated with T-Ⅰ,and group R and R+T-Ⅰ were exposed to radiation after 24 h.Cell viability and reactive oxygen species (ROS) were compared among all groups.Results T-Ⅰ attenuated radiation-induced viability decrease of urothelial cells and ROS generation in vitro.In vivo,treatment of T-Ⅰ dramatically prevented radiation-induced histological damage and reduced oxidative stress in the acute phases of RIBI,while the bladder fibrosis was significantly alleviated in the chronic phase.Conclusion T-Ⅰ plays a protective role in RIBI via its antioxidant activity.
