A primary study on the detection of CK19, CD105 and CD146 in peripheral blood for postoperative monitoring of renal carcinoma

Abstract:Objective To investigate the relationship between the three molecular markers,CK19,CD105 and CD146 and the tendency of early metastasis of renal cell carcinoma (RCC).Methods A total of 200 RCC patients who received radical nephrectomy or nephron-sparing surgery during June 2013 and April 2016 were enrolled,and 100 non-RCC patients served as controls.The expressions of CK19,CD105,and CD146 were detected by immunohistochemical staining,Western blotting,and ELISA.Results CK19 and CD105 were highly expressed in tumor tissues,but scarcely expressed in adjacent normal (or other non-renal carcinoma) tissues,with statistical difference.There was difference in the expression of CD146 between tumor tissues and adjacent normal tissues.Further studies showed no significant difference at four time points (Q1:1 day pre-operation;Q2:1 day post-operation;Q3:1 week post-operation;and Q4:1 month post-operation) in the expressions of CK19,CD105 and CD146.Subgroup analysis based on the circulating tumor cells (CTCs) showed there was no difference in the expressions of CK19,CD105 and CD146 between Q2 and Q1;there were significant differences in the expressions of CK19 and CD105 between Q3 and Q1;there were significant differences in the expressions of CK19,CD105 and CD146 between Q4 and Q1.Conclusion The ELISA test of CK19,CD105 and CD146 in peripheral blood might be used to assess whether there is an early trend of metastasis in patients with renal cell carcinoma.However,a multicenter,large sample,longer-term follow-up monitoring research is needed to support our conclusion.
