The establishment of female rats mating model

Abstract:Objective To establish the best female rats mating model. Methods Total of 30 normal female SD rats were randomly divided into three groups including the natural group(10 rats), the progesterone group(10 rats) and the castration group(rats). Rats in the progesterone group were injected with 500 ug progesterone at 4h before the mating test. Rats in the castration group whose underwent bilateral ovariectomized were injected with 20μg estradiol benzoate and 500ug progesterone at 48h and 4h respectively before mating. Rats in the natural group were as the control. Vaginal smear was performed before mating test, and then the mating test was done. Results Only 1 rat was in estrus in the natural group, successful mating rate of this group was 10%; Two rats of progesterone group were in estrus, and the successful rate was 20%; rats of castration group were all in estrus, and the successful rate was 100%. Conclusion Male sexual behavior was closely related to female estrus. Hormone-treated females castration rats were the best animal mating models.
