Gene Expression of Extracellular-Signal Regulated Protein Kinase 5 and Their MAPKK in Fetal Skin at Different Develop Mental Stages and Hypertrophic Scars

Abstract:Objective:To explore the change in gene expression of extracellular-signal regulated protein kinase 5 (erkS)and its upstream signaling molecule(mek5)in fetal skin at different developmental stages and hypertrophic scars and their potential biological significance.Methods:After morphological characteristics of skin specimens ob- tained at different developmental stages and hypertrophic scars were defined with histological methods,gene ex- pressions of erk5 and mek5 were assessed with reverse transcription-polymerase chain reaction(RT-PCR). Results:In early gestational fetal skin,genes of erk5 and reek5 were strongly expressed,while in late gestational skin and childhood skin,gene expressions of these two genes were apparently weakened(P0.05).In normal skin,gene expression level of erk5 was comparatively lower.In proliferating hypertrophic scars,mRNA content of this gene was apparently increased.In mature scars,the content of this gene transcript was 3.2 fold of that of normal skin.The level of this gene expression was significantly elevated in comparison with normal skin(P 0.05).In contrast,the levels of mek5 transcript in normal skin and hypertrophic scars in various postwound peri- ods showed no substantial change(P0.05).Conclusion:Erk5 mediating signaling pathway might be involved in regulating cutaneous development at embryonic stage and in determining cutaneous structure and function.The relative increment of gene transcription of erk5 and reek5 in younger fetal skin might be one of the reasons why cu- taneous cells proliferate rapidly and the wounds heal without scar.The activation of erk5 gene expression in hyper- trophic scars versus in normal skin might be one of the mechanisms controlling the formation of hypertrophic scars,in which the role of mek5 needs to be further investigated.
