Effects of heat stress in cancer-associated fibroblasts on proliferation and migration of colon cancer cells

Abstract:Objective:Cancer associated fibroblasts (CAFs) were treated with heat stress. The effect of heated CAFs on the proliferation and migration of colon cancer were observed.Methods:Primary cells of CAFs were extracted and cultured from colon cancer tissues. Heat-stress conditioned medium were collected from the supernatant fluid of medium in which CAFs were treated in 43 ℃ for 1 h. Control medium were collected from the supernatant fluid of medium from CAFs in 37 ℃. The study aimed to investigate the effect of heated-stress conditioned medium in SW-480. Cell counting kit-8, wound healing and Transwell assays were performed to determine whether heated-stress conditioned medium effects on cell proliferation and migration. Enzyme-linked immune sorbent assay was used to detect the expression level of interleukin-11 in the supernatant of conditioned mediums.Results:The proliferation of SW-480 cells treated by heated-stress conditioned medium was stronger in 72 h and 96 h ( t=3.915, P<0.05; t=3.866, P<0.05). The migration of SW-480 treated by heated-stress conditioned medium was also enhanced. The wound healing rate in 48 h in heated-stress conditioned medium was higher in wound healing assay [(38.9±6.2)% vs. (14.7±4.1)%, t=4.616, P<0.05], and the number of transplanted colon cancer cells co-cultured with heat stress CAFs was more than that co-cultured with control CAFs [(173±30) cells vs. (51±5) cells, t=5.591, P<0.01]. The relative absorbance of IL-11 in heat-stress conditioned medium was significantly higher than in control medium ( t=2.790, P<0.05) in the transwell co-culture system. Conclusion:Heat stress CAFs can enhance the proliferation and migration of colon cancer cells, which may be related to the paracrine of IL-11.
