Effect of new nanocomposite RGD peptide conjugated poly [LA-(Glc-Lys)]/poly (D, L-lactic acid)/nanosized hydroxyapatite/valproic acid nerve conduits on peripheral nerve regeneration in a rat model

Abstract:Objective To observe the enhancing effect of nerve regeneration on peripheral nerve defect models bridged by a new RGD peptide conjugated poly [LA-(Glc-Lys)]/Poly (D,L-lactic acid)/nanosized hydroxyapatite/valproic acid (PRGD/PDLLA/n-HAP/VPA) nanocomposite conduit.Methods In this study,30 rats were randomly divided into 3 groups (n =10):group A,PRGD/PDLLA/n-HAP/VPA;group B,PDLLA;group C:autograft.Results At 12th week after surgery,the sciatic functional index (SFI) values in groups A (-41.00±3.08) and C (-39.00 ±3.12) were significantly higher than those in group B (-73.00 ± 3.11,P < 0.05).The compound muscle action potentials (CMAPs,mV) in groups A (24.78 ± 4.05) and C (25.39 ± 5.54) were significantly higher than those in group B (14.74 ±3.09,P<0.05).The nerve conduction velocity studies (NCVs,m/s) in groups A (31.39 ±2.38) and C (31.53 ±2.11) were significantly higher than those in group B (20.07 ± 2.31,P <0.05).The number of regenerated nerve in the tube of groups A (256.00 ±6.08) and C (258.00 ± 5.49) was significantly greater than that of group B (231.00 ±5.16,P <0.05).There was no significant difference between groups A and C (P > 0.05).Conclusion These promising results illustrate that this PRGD/PDLLA/n-HAP/VPA nanocomposite conduit has great potential for clinical applications.
