Effect of neurotensin/interleukin-8/CXC chemokine ligand 1 signaling on the cancer stem-like phenotype and invasive and metastatic ability of hepatocellular carcinoma and action mechanism

Abstract:Objective To study the effecof neurotensin (NTS)/interleukin (IL)-8/CXchemokine ligand 1 (CXCL1) signaling on the cancestem-like phenotype, and invasive and metastatiability of hepatocellulacarcinom(HCC) and the possible mechanism.MethodMHCC97-H cellwere divided into three groups: normal control group, NTgroup (1 mg/L NTS), NT+ BAY11-7082 group (1 mg/L NT+ 10 μmol/L BAY11-7082).The proportion of CD133 positive cellwadetermined using flow cytometry (FCM).Enzyme linked immunosorbenassay (ELISA) waemployed to measure the levelof IL-8 and S100A12 in the supernatant.Western blotting waused to detecthe S100A12 and CXCL1 and nucleafactor-κ(NF-κB) p65 expression.Transwell assay waconducted to testhe invasive and metastatiability of the tumocells.ResultAcompared with normal control and NT+ BAY11-7082 groups, the proportion of CD133 positive cells, the contenof IL-8 and S100A12 in the supernatant, and the numbeof cellinfiltrating Transwell membrane were significantly increased, while the expression of S100A12 and CXCL1 significantly increased in the NTgroup (P <0.05).NTalso effectively promoted the subscquenNF-κnuclcatranslocation.All these effecof NTcould be converted by NF-κinhibito(BAY11-7082).Conclusion NTS/IL-8/CXCL1 signaling could regulate the cancestem-like phenotype, the expression of the S100A12, and invasive and metastatiability of MHCC97-H cellviNF-κB.
