Protective effects of electrical stimulation on mechanical stress -induced oxidative damage in mouse fibroblast L 929 cells

Abstract:Objective To explore the protective effects of electrical stimulation on mechanical stress -induced oxidative damage in mouse fibroblast L 929 cells.Methods The mouse fibroblast L 929 cells were divided into four groups : blank control group, electrical stimulation group, mechanical stress group and electrical stimulation after mechanical stress group .The cell viability and cellular senescent state were analyzed using CCK -8.Fluorometric assays were performed to detect the formation of reactive oxygen species (ROS) and mitochondrial membrane potential detection (JC-1) staining in the cells.The caspase-3 activity was detected by Caspase-3 Activity Assay Kit.Results The cell viability in mechanical stress group decreased significantly , intracellular ROS increased, mitochondrial membrane potential decreased , and Caspase-3 enzyme activity increased.The differences were statistically significant (P<0.05).Compared with mechanical stress group , the cell viability in electrical stimulation after mechanical stress group increased, the mitochondrial membrane potential increased , the intracellular ROS content decreased , and the Caspase -3 enzyme activity weakened.The differences were statistically significant ( P<0.05).Conclusion Electrical stimulation can maintain cell viability, reduce cell apoptosis, reduce intracellular ROS and Caspase -3 activity.Therefore,the electrical stimulation may be a potential method for the prevention and treatment of mechanical stress -induced cell injury.
