Significance of Detecting Disseminated Cancer Cells in Ascites or Peritoneal Lavage Fluid of Ovarian Cancer Patients with Survivin Mrna

Abstract:Objective:To explore the significance of detecting disseminated cancer cells in ascites or peritoneal lavage fluid of ovarian cancer patients with Survivin mRNA.Methods:Ascites or peritoneal washes were obtained from 50 ovarian cancer patients during laparotomy.The Survivin mRNA levels in these materials were determined using reverse-transcriptase polymerase chain reaction (RT-PCR).Conventional cytological examinations were also performed.All patients have been followed up for 2 years.Results:The positive rate of Survivin mRNA was 58%,which was higher than that of cytological method (38%).The combination of RT-PCR and cytological examination yielded positive result in 62% (31/50) patients with ovarian cancer.Expression of Survivin mRNA was significantly correlated with cancer stage classification (P0.05),but negatively related with pathological type,histological grade,and lymph node metasatasis (P0.05).9 patients had clinical evidence of peritoneal metastasis in two years.Two cases with negative results of conventional cytology were positive of Survivin mRNA.Conclusions:Survivin specific RT-PCR targeting micro-metastasis on ascites or peritoneal washes is more sensitive than conventional cytology.Combination of these two methods will produce important significance for clinical stage and evaluation of prognosis in ovarian cancer.
