Thymidine Phosphorylase and Angiogenesis in Breast Carcinoma

Abstract:Breast cancer is angiogenesis-dependent, thymidine phosphorylase, a kind of pro-angiogenic factor which expresses higher in cancer tissue than that in surrounded normal tissue plays critical role in the angiogenic switch of breast cancer. The mechanism is related primarily to 2-deoxy-D-ribose(2-dDR), the product of thymidine by TP specially. 2-dDR could promote the angiogenesis by many ways such as, the formation of blood vessel-like structure via inducing the concentration-dependent endothelial migration, the increased anti-apoptosis activity due to the protective role of CO induced by the higher expression of HO, and the release of pro-angiogenic factor,e.g. VEGF, IL-8, MMP-1 induced by oxidative stress. The expression of TP could be induced by low oxygen level and pH in microenvironment, and many cytokines such as IL-1, TNF-α, IFN-γ could increase the level of TP and its enzymatic activity. The anti-angiogenesis therapy targeted to TP is individual and knowing the expression of TP has a guide role in judging the prognosis, therapy response and establishing reasonable treatment strategy in order to improve the prognosis of the patients.
