The Expression of Nucleolin and VEGF in Colon Cancer and Their Role in Metastasis

Abstract:Objective: To research the expression of Nucleolin and VEGF in colon cancer as well as the relation-ship between Nucleolin, VEGF and clinicopathological features of colon cancer. Methods:Immunohistochemistry was used to detect the expression of Nucleolin and VEGF protein in 60 cases of colon cancer and to analyze the correlation between these two parameters and the pathological features of colon cancer. Results: VEGF was positively expressed in the cyto-plasm in normal colon tissue, colon cancer and metastatic colorectal cancer tissues with the scores of 3. 85 ± 0. 47,4. 93 ± 0. 40 and 6. 80 ± 0. 70, respectively with significant difference (P<0. 05). Nucleolin was positively expressed in the nu-cleus, cytoplasm and nuclear membrane in normal colon tissue, colon cancer and metastatic colorectal cancer tissues with the total scores of 2. 65 ± 0. 47,3. 43 ± 0. 55,4. 03 ± 0. 65, respectively. There was statistically significant difference of Nu-cleolin overall expression levels between normal colon tissue and colon cancer tissue (P <0. 05), while no significant difference was found between colon cancer and metastatic colon cancer (P>0. 05). Nucleolin total protein levels in in situ colon cancer , lymph node metastasis and liver metastasis of colon cancer showed no significant difference (P>0. 05). However, in metastatic colon cancer, nuclear staining of Nucleolin was weaker, cytoplasm and membrane staining was en-hanced. VEGF expression levels in lymph node metastasis and liver metastasis of colon cancer tissues were significantly higher than that in in situ colon cancer (P=0. 02), which was especially highly expressed in liver metastasis. Conclu-sion:The translocation of Nucleolin protein from nuclear to membrane may be associated with the metastasis of the colon cancer. VEGF may be involved in this process. Nucleolin will be expected to become an effective target for biotherapy <br> for metastatic colon cancer.
