Abstract:Objective: To observe the inhibitory effect of point-mutatedhuman recombinant interferon-β on the growth of human tumor cells in vivo and in vitro. Methods: Human colon carcinoma cell line LoVo and human lung carcinoma cell line A549 were cultured in PRMI 1640 supplemented with human recombinant IFN-β under the indieated concentrations (200 IU/ml, 1 000 IU/ml, 5 000 IU/ml) for 24 h respeetively. These cells were then cultured in RPMI 1640 with 0.3% gelose for 10 to 14 days and the colonies were counted. LoVo cells and A549 cells were also inoculated subcutaneous in BALB/c nude mice. Three days later, these mice were treated with point-mutated human recombinant IFN-β(2×105 IU/kg, 1×106 IU/kg or 5×106 IU/kg respeetively)once every day for 10 days. Then the mice were killed and the tumors were isolated and weighted. Results: The number of colonies formed by the tumor cells treated with mutated recombinant IFN-β was much lower than that of colonies formed by tumor cells without treatment (0.05>P>0.01). But there was no difference between the mice treated with the recombinant IFN-β statistically (P>0.05). The weight of tumors from the mice treated with IFN-β decreased significantly compared with the mice without treatment. No difference was observed (P>0.05) compared to the group of standard. Conclusion: The growth of some human tumor cells can be inhibited by point-mutated recombinant IFN-β not only in vivo but also in vitro.
