A Research of Brain Glucose Metabolism Characteristics of Alzheimer’s Disease, Lewy Body Dementia and Parkinson’s Disease with Dementia Based on Modular Analysis of Metabolic Networks

Abstract:ABSTRACT Aim The network structure of human brain is composed of interconnected modules. The modular analysis of brain network based on functional brain imaging can reflect the attributes and functional connections of brain network. This study aimed to explore the characteristics and similarities together with differences of three dementia subtypes Alzheimer’s disease ( AD) , dementia with Lewy body ( DLB) and Parkinson’s disease with dementia ( PDD) brain network module parameters, and to find disease-related specific brain glucose metabolism network module parameters. Methods 18 F-FDG PET imagings of resting state in AD, DLB, PDD group and healthy control group in Huashan Hospital Affiliated to Fudan University were retrospectively analyzed. Modular parameters of glucose metabolism network in four groups of subjects were obtained by modular analysis. The corresponding connection changes of brain regions were obtained by seed point correlation analysis. The similarities and differences of brain network connectivity between the four groups were acquired. Results When the sparse threshold was 15% , the network of the control group and AD group were composed of four modules, PDD group was composed of six modules, and DLB group was composed of 10 modules. With the right thalamus as the seed point, it was found that in AD group, strengthened connections with right thalamus were focused on temporal gyrus and subcortical gyrus, while weakened connection regions were mainly located in occipital gyrus and left frontal gyrus. In PDD group, strengthened connections with right thalamus were focused on frontal gyrus and subcortical gyrus, while weakened connection regions were located in occipital gyrus, temporal gyrus and left parietal gyrus. In DLB group, strengthened connections with right thalamus were focused on left frontal gyrus and subcortical gyrus, while weakened connection regions were mainly located in right occipital gyrus and parietal gyrus. Conclusion The metabolic networks of the three different subtypes of dementia had different modularity characteristics which might suggest different pathogenesis of these diseases.
