Effect of fixatives on cytochrome oxidase histochemistry

Abstract:Cytochrome oxidase (CO) is a terminal enzyme of mitochondrial respiratory chain and a vital component of cellular energy production and transduction. In the central nervous system, regional CO activity provides an index for local tissue energy metabolism, which is closely coupled to neuronal depolarizing activities, in particular of synaptic and spontaneous types. CO histochemistry has been widely used to demonstrate CO activity. For successful CO histochemistry, proper fixation is crucial. Compromise has to be made between the quality of tissue fixation and the preservation of enzyme activity. Usually, a mixture of paraformaldehyde and glutaraldehyde is used. The present study was aimed to work out the best combination of these two fixatives for both light and electron microscopic CO histochemistry. It was found that a mixture of 1% paraformaldehyde and 2% glutaraldehyde serves the purpose best in the areas of the central nervous system of the adult rat we investigated.
