Expert Opinion on Therapeutic Targets

Abstract:About a decade ago, the death factor Fas ligand (FasL) was identified as the natural trigger of Fas/CD95-dependent apoptosis and as an inducer of Fas-dependent activation-induced cell death. Meanwhile, it is known that this molecule not only contributes to target cell lysis in the immune system but also to the establishment of immune privilege and tumour survival. Because delivering a specific antiproliferative signal to T lymphocytes is of major biomedical interest, the FasL/Fas system has gained much attention over the last few years. However, only recently it became evident that the biology of FasL is more complex than initially anticipated. FasL displays a complex pattern of inducible and constitutive expression associated with a number of different functions as a death factor or a co-stimulatory/accessory molecule in lymphocyte activation. Thus, side effects are likely to occur following systemic administration of, for example, anti-FasL medication, not only because of the constitutive FasL expression on cells within immune privileged tissues and vascular endothelium. In addition, FasL comes in different forms: as a surface molecule, as a protease-shed soluble variant or secreted in vesicles. Because increased levels of soluble FasL (sFasL) have been determined in various immunological and non-immunological diseases, it has been suggested that sFasL might serve as a prognostic or diagnostic marker even though the pathophysiological cause for its enhanced production is hardly known in most cases. This review summarises the current facts and ideas about the clinical and pharmacological potential of FasL and sFasL as targets for therapeutic interventions.

Abstract:AbstractFas ligand (FasL, CD95L) is the key death factor of receptor‐triggered programmed cell death in immune cells. FasL/Fas‐dependent apoptosis plays a pivotal role in activation‐induced cell death, termination of immune responses, elimination of autoreactive cells, cytotoxic effector function of T and NK cells and the establishment of immune privilege. Dysregulation of FasL‐expression has severe impact on the maintenance of immune homeostasis and defense and is often associated with pathophysiology. Moreover, it was recently shown that FasL serves as a costimulatory receptor during T cell activation. However, it is still not known whether FasL‐ligation induces a ‘direct’ signal or whether the regulation of activation signals is a more ‘indirect’ result of alterations in intracellular protein‐protein‐interactions. In the latter scenario, the proline rich domain of FasL was shown to be required for a directed transport and localization of FasL. Over the past years, potential FasL‐associating molecules were identified which mostly bind via Src homology 3 or WW domains to the proline rich domain of FasL. This review summarizes facts and hypotheses about these FasL‐interacting proteins. It reflects their potential role in reverse signal transduction and FasL biology in T lymphocytes and other cell types.

Abstract:Fas ligand (FasL) is critical for maintaining immune privilege status of central nervous system in physiological conditions.FasL binds Fas via extrinsic pathway mediated ischemic penumbra neuronal apoptosis after the onset of cerebral ischemia.In addition,FasL is involved in the regulation of T lymphocyte subsets by inducing the expression of inflammatory cytokines and chemokines,promoting intracerebral microglia activation and peripheral neutrophil infiltration,and thus aggravating immune inflammatory response after stroke.FasL-Fas system has a certain role for promoting the recovery of neural function during the convalescence of ischemic stroke.Taking FasL-Fas system as a target,it may become a new way for brain protection treatment of ischemic stroke.
Activation‐dependent FasL Expression in T Lymphocytes and Natural Killer Cells

Abstract:AbstractIn cytotoxic T cells and Natural Killer cells, the death factor FasL is stored in association with secretory granules. Only upon activation, these vesicles are transported to the cytotoxic immunological synapse and FasL is expressed on the cell surface. Upon activation of T cells and NK cells with phorbol ester and calcium ionophore, we observed a biphasic expression of FasL in all lymphocyte subsets tested. The first peak was seen after 10−15 minutes of stimulation and was followed by a reduction of expression to baseline level before the second peak was reached at about two hours. Using inhibitors of protein biosynthesis (cycloheximide), vesicular transport (brefeldin A and monensin), actin polymerization (latrunculin A and cytochalasin D) and metalloproteases (GM 6001 and phenanthrolin), the regulation of FasL expression was analyzed in detail. We demonstrate that the first wave of expression is due to an actin‐dependent mobilization of preformed FasL whereas the second phase of expression requires de novo synthesis. The observed expression patterns might have implications for the development of therapeutic strategies that target FasL as an immunomodulatory protein.

Abstract:Compared with other cells,more high level of Fas and Fas ligand(FasL) expression have been detected in immunocytes.Fas/FasL-mediated cell apoptosis and prolife-ration are intimately associated with function development of immunocytes,which play an important role in asthmatic immunotolerance and regulatory abnormity,Th1/Th2 imbalance,airway inflammation,mucus generation,airway hyperresponsiveness and remodeling,leading to a breakthrough hopefully in asthmatic therapy.Still,in Fas/FasL signal transduction,different factors form a complex regulatory network.So,further research regarding the mechanisms by which the expression of Fas and FasL is regulated,will be developed to seek a new way in the treatment of asthma.

Abstract:Background: Regulatory DCs are important in tolerance maintenance, whereas the mechanisms for maintaining their immunosuppressive function in immune microenvironment remain unclear. Results: Fas signal enhances the immunosuppressive function of splenic stroma-educated regulatory DCs via the ERK/β-catenin pathway. Conclusion: Fas signal, at least from activated T cells, enhances regulatory function of regulatory DCs. Significance: This study provides new mechanistic insight for immune homeostasis. Dendritic cells (DCs) play important roles in the initiation of immune response and also in the maintenance of immune tolerance. Now, many kinds of regulatory DCs with different phenotypes have been identified to suppress immune response and contribute to the control of autoimmune diseases. However, the mechanisms by which regulatory DCs can be regulated to exert the immunosuppressive function in the immune microenvironment remain to be fully investigated. In addition, how T cells, once activated, can feedback affect the function of regulatory DCs during immune response needs to be further identified. We previously identified a unique subset of CD11bhiIalow regulatory DCs, differentiated from mature DCs or hematopoietic stem cells under a stromal microenvironment in spleen and liver, which can negatively regulate immune response in a feedback way. Here, we show that CD11bhiIalow regulatory DCs expressed high level of Fas, and endothelial stromal cell-derived TGF-β could induce high expression of Fas on regulatory DCs via ERK activation. Fas ligation could promote regulatory DCs to inhibit CD4+ T cell proliferation more significantly. Furthermore, Fas ligation preferentially induced regulatory DCs to produce IL-10 and IP-10 via ERK-mediated inactivation of GSK-3 and subsequent up-regulation of β-catenin. Interestingly, activated T cells could promote regulatory DCs to secrete more IL-10 and IP-10 partially through FasL. Therefore, our results demonstrate that Fas signal, at least from the activated T cells, can promote the immunosuppressive function of Fas-expressing regulatory DCs, providing a new manner for the regulatory DCs to regulate adaptive immunity.

Abstract:In order to evaluate the importance of FasL in the apoptosis of lymphocytes in experimental autoimmune encephalomyelitis (EAE),a mouse model of multple selerosis (MS) EAE (MS-EAE) that was induced by inoculation with myelin basic protein (MBP) and complete Freund's adjuvant into mice was constructed in this study,and the expressions of Fas and FasL on the surface of lymphocytes in EAE were analyzed thereby. It was found that the expressions of Fas and FasL as well as that of IL-4 and IL-10 of the lymphocytes in EAE were to be higher than that of the controls,and FasL could induce apoptosis of lymphocytes in a dose-dependent manner on the dosage of 1～100 ng/ml. The Fas-induced apoptosis could be blocked partially by the addition of specific monoclonal antibody against FasL. These results indicate that Fas-FasL interaction plays an important role in the pathogenesis of autoimmune disease.

Abstract:As a member of the tumor necrosis factor (TNF)/nerve growth factor (NGF) receptor superfamily, Fas (CD95) plays an important role in apoptosis and immunoregulation. Once Fas binds with its ligand, known as Fas ligand, the cells expressing Fas goes to program cell death or apoptosis. Apoptosis mediated by Fas-FasL involves in the clonal deletion of T cell and B cell in thymus and bone marrow at the early stage of the cell’s differentiation. CTL and NK kill their target cells by Fas and FasL interaction. So the Fas and FasL are very important elements in maintaining immune system homeostasis of the human body and involved in autoimmune disorders, tumor development and acquired immune deficiency syndrome.

Abstract:Originally identified as a cell surface receptor that triggered the death of lymphocytes and tumor cells, it is now recognized that Fas (also known as CD95 or Apo-1) has distinct functions in the life and death of different cell types in the immune system. Fas signaling may also be involved in T cell costimulation and proliferation. Although Fas deficiency in humans and mice predisposes them towards systemic autoimmunity, Fas-FasL interactions can also facilitate organ-specific immunopathology. Proximal signaling by Fas and related receptors depends on subunit preassembly, which accounts for the dominant-negative effect of pathogenic receptor mutants and natural splice variants.
Targeted mutation of Fas ligand gene attenuates brain inflammation in experimental stroke

Abstract:Inflammation is an important contributing mechanism in ischemic brain injury. The current study elucidates a previously unexplored role of Fas ligand (FasL) in post-stroke inflammatory responses that is independent of its well-known effect in triggering apoptosis. Focal cerebral ischemia was induced for 2 h by right middle cerebral artery occlusion (MCAO) in FasL mutant (gld) and wild-type mice. FasL mutation profoundly reduced brain damage and improved neurological performance from 6 to 72 h after ischemic stroke. The production of inflammatory cytokines in the brain was attenuated in gld mice after ischemia in the absence of dramatic change in inflammatory cell apoptosis. FasL mutation attenuated the recruitment of peripheral inflammatory cells (neutrophil) and inhibited the activation of residential glial cells (microglia and astrocyte). FasL mutation reduced CD8(+) T cells and turned the Th1/Th2 balance towards Th2 in the brain and peripheral blood after cerebral ischemia. In contrast to cerebral ischemia, the molecular and cellular inflammatory changes induced by intracerebroventricular injection of lipopolysaccharide (LPS) were also attenuated in gld mice. Moreover, the soluble FasL (sFasL) and phospho-SAPK/JNK were decreased in gld mice, suggesting that the inflammatory role of FasL in experimental stroke might relate to sFasL and the c-Jun N-terminal kinase (JNK) signaling pathway. Taken together, our data suggest a novel role of FasL in the damaging inflammatory responses associated with cerebral ischemia. Neutralization of FasL may be a novel therapeutic strategy to suppress post-stroke inflammation and improve the long-term outcomes of stroke.

Abstract:BackgroundActivated microglia play a vital role in neuroinflammation in the central nervous system (CNS), which is associated with the pathogenesis and the progression of neurological diseases. Interferon regulatory factor 5 (IRF5) has been well established participating in inflammatory responses and is highly expressed in M1 macrophage in the periphery, the role of which in the CNS remains elusive.MethodsLipopolysaccharide (LPS) was employed to induce neuroinflammation. Down-regulation of IRF5 in C57/BL6 mice and BV2 microglial cells were achieved by IRF5 siRNA transfection. The levels of pro-inflammatory cytokines were evaluated by ELISA and quantitative real-time PCR. The expression levels of IRF5 were examined by immunofluorescence and Western blot.ResultsLPS induced significantly elevated expression of IRF5 in mouse brain, which co-localized with CD11b-positive microglia. Down-regulation of IRF5 quenched the pro-inflammatory responses. The levels of pro-inflammatory cytokines TNF-α, IL-1β, and IL-6 were up-regulated at 4 h after LPS treatment, which were significantly down-regulated with the knockdown of IRF5. LPS-induced pro-inflammatory responses were transient, which were comparable to control group at 24 h after LPS treatment. However, LPS did not up-regulate the expression of IRF5 in BV2 microglial cells, indicating that LPS-induced inflammation in BV2 cells does not involve IRF5 signaling.ConclusionsIRF5 mediates the inflammatory responses in the CNS, which might serve as a therapeutic target for CNS inflammatory diseases. LPS-induced inflammation does not involve IRF5 signaling in BV2 microglia.
Fas/FasL Contributes to HSV-1 Brain Infection and Neuroinflammation

Abstract:The Fas/FasL pathway plays a key role in immune homeostasis and immune surveillance. In the central nervous system (CNS) Fas/FasL is involved in axonal outgrowth and adult neurogenesis. However, little is known about the role of the Fas/FasL pathway in herpes encephalitis. In this study, we used a neuropathogenic clinical strain of herpes simplex virus type 1 (HSV-1) to explore infection-induced inflammation and immune responses in the mouse brain and the role of Fas/FasL in antiviral CNS immunity. HSV-1 CNS infection induced the infiltration of Fas- FasL-bearing monocytes and T cells in the brain and also to an up-regulation of Fas and FasL expression on resident astrocytes and microglia within infected sites. Upon infection, Fas- and FasL-deficient mice (lpr and gld) were partially protected from encephalitis with a decreased morbidity and mortality compared to WT mice. Fas/FasL deficiency promoted cell-mediated immunity within the CNS. Fas receptor stimulation abrogated HSV-1 induced activation and inflammatory reactions in microglia from WT mice, while lack of Fas or FasL led to a more pronounced activation of monocytes and microglia and also to an enhanced differentiation of these cells into a pro-inflammatory M1 phenotype. Furthermore, the specific immune system was more efficient in Fas- and FasL-deficient mice with significantly higher numbers of infiltrating HSV-1-specific cytotoxic T cells in the brain. Our data indicate that the Fas/FasL pathway leads to excessive neuroinflammation during HSV-1 infection, which is associated with a diminished anti-viral response and an excessive neuroinflammation.

Abstract:Fas antigen(Fas)is a type Ⅰ membrane protein belonging to the tumor necrosis factor receptor(TNFR)/nerve growth factor receptor(NGFR)super family.Cross-linking of Fas with its ligand,FasL,or its competitive antibody can induce cell apoptosis.The signal transduction and regulatory mechanisms of Fas plays an important role in keeping self balance in organism and the development of many deseases.It has also been proved that Fas signaling plays a critical role in pulmonary interstitial fibrosis in recent years.The recognition of this mechanism may provide a new method in the therapy of current -pulmonary interstitial fibrosis.
Effect of HIV-1 Infection on Apoptosis of CD4+T Lymphocytes Mediated by Fas

Abstract:OBJECTIVE:To study the pathogenic role of Fas/CD95 in HIV-1 infection subjects, and to investigate the effects of HIV on plasma levels of sFas and the expression of CD95 on different CD4(+) T lymphocyte subpopulations.METHODS:Four-color flow cytometry was used to determine the expression of CD95, CD45RO, CD45RA on CD4(+ )T lymphocyte in peripheral blood from HIV-1 infection subjects and serum Fas levels were quantified by enzyme-linked immunosorbent assay (ELISA).RESULT:Compared with healthy controls, serum Fas levels were significantly increased (P<0.05) in HIV group and positively correlated with the disease progress. The expression of CD95 on naive T-lymphocyte subsets was increased whereas that on memory T-lymphocyte subsets was decreased.CONCLUSION:Fas plays an important role in the deletion of CD4(+) T-lymphocyte during HIV-1 infection. Further understanding of the relationship between Fas/CD95 and CD45RO/CD45RA may help to predict the progression of the disease and the clinical outcome.
FasL Incapacitation Alleviates CD4+ T Cells-Induced Brain Injury Through Remodeling of Microglia Polarization in Mouse Ischemic Stroke.

Abstract:Inflammation responses involving the crosstalk between infiltrated T cells and microglia play crucial roles in ischemia stroke. Recent studies showed that Fas ligand (FasL) mutation could reduce post-stroke T cell invasion and microglia activation. In this study, we demonstrated that CD4+ T cells could induce M1 microglia polarization through NF-κB signaling pathway, whereas FasL mutant CD4+ T cells significantly reversed this effect. Besides, Th17/Treg cells balance was skewed into Treg cells after FasL mutation. In addition, conditioned medium from co-culture of FasL mutant CD4+ T cells and microglia could alleviate neuronal injury. Collectively, FasL incapacitation could alleviate CD4+ T cells-induced inflammation through remodeling microglia polarization, suggesting a therapeutic potential for control of inflammation responses after ischemic stroke.

Abstract:Background: Inflammation is closely implicated in the pathogenesis and outcome of ischemic stroke. Although crosstalk between microglia and T cells has been studied in CNS diseases such as Parkinson’s disease, yet few research has focused this relationship on stroke, and no definite mechanism has been proposed to explain the relationship. In this study, we discussed the crosstalk between microglia and T cells and the probable role of FasL in the progress. Methods: Primary microglia were extracted from the cortexes of new born C57BL/6J (B6) mice while primary T cells were extracted from spleens of adult B6 mice. MCAO and OGD model were used to mimic cerebral ischemic injury in vivo and in vitro respectively. T cell chemotaxis assay was determined by transwell system. Cell markers in vivo were detected by immunofluorescence. The expression of cytokines were detected by Q-PCR. The amount of soluble FasL into the cultured media were measured by ELISA. Results: In vivo , T cells infiltrated into the ischemic lesions and aggregated around microglia within 24 h after MCAO. The expression of FasL in microglia was also up-regulated after MCAO. However, in FasL mutant ( Gld ) mice, the infiltration and aggregation of T cells were induced. In vitro , when coculturing primary T cells with primary microglia, crosstalk also existed between them morphologically after OGD. Furthermore, proinflammatory cytokines such as IL-1β, IL-6 were increased and anti-inflammatroy cytokines such as IL-10 were reduced by Q-PCR. However, the crosstalk wasn’t obvious in Gld primary cocultured cells. On the other hand, soluble FasL was increased in the supernatant of activated microglia by ELISA. In chemotaxis assay, T cells were chemotactic to supernatant of activated microglia while the effect was weakened after inhibiting FasL or MCP-1, which indicating that soluble FasL released by microglia may also play an important role in chemoattracting T cells. Conclusions: These data suggest that an inflammatory crosstalk take place between activated microglia and T cells after cerebral ischemia, in which FasL may play an important part. Microglia may chemoattract periphery T cells by releasing soluble FasL after ischemic injury.

Abstract:AIM: To investigate the role of soluble Fas ligand in autoimmune diseases.METHODS: RT-PCR was performed to amplify sFasL cDNA from the total RNA extracted from activated human peripheral blood lymphocytes. DNA fragments were cloned into PCR vector. After sequenced, sFasL gene fragments were inserted into pQE-31 vector and expressed in E. Coli M15 respectively. Proteins were purified through affinity chromatography column with ligand of 6xHis tag and identified by SDS-PAGE and Western blot. Mice were immunized with sFasL protein and specific anti-serum was harvested 6 wk after immunization. Monoclonal anti-human FasL antibody was made from the immunized mice. Serum level of sFasL in different patients was detected using anti-FasL antibodies from the immunized mice.RESULTS: The protein expressed was 24 ku by SDS-PAGE electrophrosis. The protein was specially bound to antihuman FasL antibody by Western blot analysis. The sFasL protein could induce Jurket cell apoptosis in vitro. The concentration of serum sFasL in patients with autoimmune diseases was higher than that in normal individuals. sFasL could reduce arthritis in collagen induced arthritis (CIA) mice model by subcutaneous injection.CONCLUSION: sFasL may be involved in either induction of apoptosis or autoimmune diseases. Furthermore, sFasL may have potential application in treatment of autoimmune diseases.
Fasl Expression On Human Nucleus Pulposus Cells Contributes To The Immune Privilege Of Intervertebral Disc By Interacting With Immunocytes

Abstract:The mechanisms of immune privilege in human nucleus pulposus (NP) remain unclear. Accumulating evidence indicates that Fas ligand (FasL) might play an important role in the immune privilege of the disc. We aimed for addressing the role of FasL expression in human intervertebral disc degeneration (IDD) and immune privilege in terms of the interaction between NP cells and immunocytes via the FasL-Fas machinery. We collected NP specimens from 20 patients with IDD as degenerative group and 8 normal cadaveric donors as control. FasL expression was detected by qRT-PCR, western blotting and flow cytometry (FCM). We also collected macrophages and CD8(+) T cells from the peripheral blood of patients with IDD for co-cultures with NP cells. And macrophages and CD8(+) T cells were harvested for apoptosis analysis by FCM after 2 days of co-cultures. We found that FasL expression in mRNA, protein and cellular resolutions demonstrated a significant decrease in degenerative group compared with normal control (p<0.05). FCM analysis found that human NP cells with increased FasL expression resulted in significantly increased apoptosis ratio of macrophages and CD8(+) T cells. Our study demonstrated that FasL expression tends to decrease in degenerated discs and FasL plays an important role in human disc immune privilege, which might provide a novel target for the treatment strategies for IDD.

Abstract:Fatty acid metabolism, particularly fatty acid synthesis, is a very important cellular physiological process in which nutrients are used for energy storage and biofilm synthesis. As a key enzyme in the fatty acid metabolism, fatty acid synthase (FASN) is receiving increasing attention. Although previous studies on FASN have mainly focused on various malignancies, many studies have recently reported that FASN regulates the survival, differentiation, and function of various immune cells, and subsequently participates in the occurrence and development of immune-related diseases. However, few studies to date systematically summarized the function and molecular mechanisms of FASN in immune cell biology and related diseases. In this review, we discuss the regulatory effect of FASN on immune cells, and the progress in research on the implications of FASN in immune-related diseases. Understanding the function of FASN in immune cell biology and related diseases can offer insights into novel treatment strategies for clinical diseases.
